Abstract

Pengaruh penambahan bakteri Ralstonia pickettii terhadap biodegradasi DDT oleh jamur pelapuk putih Pleurotus eryngii telah diteliti. R. pickettii ditambahkan ke dalam 10 mL kultur P. eryngii masing-masing sebanyak 1, 3, 5, 7 dan 10 ml (1 ml ≈ 1,337 x 109 sel). Bakteri R. pickettii menghasilkan biosurfaktan untuk meningkatkan kelarutan DDT sehingga DDT dapat lebih mudah terdegradasi. Analisis degradasi DDT menggunakan HPLC dan Kromatografi GCMS. Degradasi DDT tertinggi ditunjukkan pada penambahan 7 mL bakteri R. pickettii dengan jumlah degradasi sebesar 78,23 % sedangkan jumlah degradasi DDT terendah terjadi pada penambahan bakteri 5 ml dengan jumlah degradasi sebesar 36,17 %. Metabolit produk yang dihasilkan adalah DDE [1,1-dichloro-2,2-bis(p-chlorophenyl)ethyene] dan DDD [1,1-dichloro-2,2-bis(p-chlorophenyl)ethane]. =========================================================== Effect of addition of bacteria Ralstonia pickettii on biodegradation of DDT by white-rot fungus Pleurotus eryngii had been investigated. R. pickettii was added into 10 mL P. eryngii culture at 1, 3, 5, 7 and 10 ml (1 ml ≈ 1,337 x 109 cells). R. picketti bacteria produced biosurfactant to increased solubility DDT so that DDT could be eassier degraded. DDT degradation analysis used HPLC and Chromatography GCMS. The addition of 7 ml of R. pickettii showed the highest DDT degradation about 78,23 %, while the addition of 5 mL of R. picketii showed the lowest one about 36,17 %. The type of metabolic product formed is DDE [1,1-dichloro-2,2-bis(pchlorophenyl) ethyene] and DDD [1,1-dichloro-2,2-bis(pchlorophenyl) ethane].
