Abstract
Diabetes mellitus tipe 2 (T2DM) yang disebabkan oleh resistensi insulin merupakan kasus diabetes yang paling banyak terjadi dan lebih dari 90% kasus diabetes mellitus merupakan T2DM. Strategi untuk pengobatan dan pengendalian T2DM dapat dilakukan dengan cara penghambatan enzim α-glucosidase. Kandungan steroid Withangulatin A pada Physalis angulata dikenal memiliki potensi sebagai inhibitor α-glucosidase. Penggunaan metode maserasi yang dilakukan pada suhu kamar dan metode refluks yang dilakukan pada suhu tinggi memengaruhi penarikan senyawa yang diinginkan pada tahap ekstraksi. Pengujian tentang pengaruh metode ekstraksi yang berbeda pada Physalis angulata terhadap penghambatan α-glucosidase dilakukan secara in vitro dengan standar acarbose sebagai inhibitor α-glucosidase dan pNPG (p-Nitrophenyl-a-D-glucopyranosie) sebagai substratnya. Nilai inhibisi kemudian dianalisis menggunakan uji ANOVA one-way yang menunjukkan signifikasi 0.000 (p < 0.05). Uji Post Hoc Tukey menunjukkan hasil bahwa rata-rata aktivitas inhibisi terbagi menjadi tiga kelompok yang berbeda dengan nilai inhibisi tertinggi berada pada sub-fraksi W.Bersih hasil ekstraksi refluks sebesar 85.30% ± 0.01. Persentase nilai inhibisi dari 6 sub-sub fraksi refluks berkisar antara 4.02% hingga 85.30%, sedangkan persentase nilai inhibisi 4 sub-sub fraksi maserasi berkisar antara 4.79% hingga 6.98%.

Insulin resistance-induced Type 2 diabetes mellitus (T2DM) is the most prevalent form of diabetes, accounting for over 90% of all diabetes mellitus cases. The inhibition of the α-glucosidase enzyme represents a viable approach for the management and containment of T2DM. The potential of Withangulatin A, a steroid found in Physalis angulata, as an α-glucosidase inhibitor has been identified. The selection of extraction method, specifically maceration at ambient temperature and reflux at elevated temperature, can impact the efficacy of the desired compound withdrawal during the extraction process. In vitro experiments were conducted to investigate the impact of various extraction techniques on the inhibition of α-glucosidase in Physalis angulata. The standard acarbose was employed as an α-glucosidase inhibitor, while pNPG (p-Nitrophenyl-a-D-glucopyranosie) was used as its substrate. The inhibition value was subjected to analysis using the one-way ANOVA test, revealing a significant result of 0.000 (p <0.05). The results of the Post Hoc Tukey test indicated that the mean inhibitory activity was segregated into three distinct groups. The W.Bersih sub-fraction obtained from reflux extraction exhibited the highest inhibition value of 85.30% ± 0.01. The inhibitory effects of the six reflux sub-sub-fractions were observed to range from 4.02% to 85.30%, whereas the inhibitory effects of the four maceration sub-fractions were found to range from 4.79% to 6.98%.
