Abstract
Salah satu jenis kanker yang paling banyak menyerang wanita adalah kanker payudara. Sel kanker payudara subtipe MDA-MB-231 memiliki karakteristik reseptor esterogen (ER), progesteron (PR), dan esterogen manusia (HER) negatif dan overekspresi reseptor folat (FR). Keberadaan FR di membran sel kanker dapat dijadikan target sistem penghantaran obat kanker karena FR dapat mengenali asam folat. Trisindolina-5 merupakan senyawa bahan alam laut yang diperoleh dari spons laut Callyspongia siphonella dan merupakan senyawa sintesis derivat dari Trisindolina-1. Trisindolina-5 memiliki aktivitas antikanker terhadap sel kanker MDA-MB-231, HT-29, OVCAR-3, dan MM.1S, namun masih bersifat toksik terhadap sel 293T (non-kanker), sehingga dilakukan enkapsulasi dengan kitosan/TPP dan konjugasi asam folat (TFNP) untuk meningkatkan efisiensi penghantaran. Fabrikasi TFNP menghasilkan efisiensi enkapsulasi 95,863±0,37%, diameter hidrodinamik (Dh) 400,9±29,05 nm, indeks polidispersitas (PDI) 0,362±0,05, dan potensial zeta (ZP) 34,34±2,92 mV. Spektrum FTIR TFNP juga menunjukkan karakter puncak yang dimiliki oleh kitosan, asam folat, TPP, dan Trisindolina-5. Uji pengambilan seluler TFNP oleh sel kanker dan nonkanker lebih tinggi dibandingkan Trisindolina-5 dan sel kanker dapat mengambil lebih banyak TFNP daripada sel nonkanker. Uji sitotoksik dari TFNP terhadap sel MDA-MB-231 dan sel 293T bersifat nontoksik pada kedua cell line dibandingkan dengan Trisindolina-5 yang bersifat toksik. Penargetan reseptor folat oleh TFNP memberikan hasil yang signifikan terhadap sel MDA-MB-231 setelah inkubasi 48 jam. TFNP mampu memicu kematian sel pada konsentrasi terendah dari uji sitotoksik yang dilakukan.

One of the most common types of cancer affecting women is breast cancer. MDA-MB-231 subtype breast cancer cells are characterized by negative esterogen (ER), progesterone (PR), and human esterogen (HER) receptors and overexpression of folate receptor (FR). The presence of FR in cancer cell membranes can be used as a target for cancer drug delivery systems because FR can recognize folic acid. Trisindolina-5 is a marine natural compound obtained from the marine sponge Callyspongia siphonella and is a synthetic compound derived from Trisindolina-1. Trisindolina-5 has anticancer activity against MDA-MB-231, HT-29, OVCAR-3, and MM.1S cancer cells, but is still toxic to 293T (non-cancer) cells, so it was encapsulated with chitosan/TPP and folic acid conjugate (TFNP) to improve delivery efficiency. TFNP fabrication resulted in encapsulation efficiency of 95.863±0.37%, hydrodynamic diameter (Dh) of 400.9±29.05 nm, polydispersity index (PDI) of 0.362±0.05, and zeta potential (ZP) of 34.34±2.92 mV. The FTIR spectrum of TFNP also showed peaks characteristic of chitosan, folic acid, TPP, and Trisindolina-5. The cellular uptake of TFNP by cancer and non-cancer cells was higher than that of Trisindolina-5 and cancer cells could take up more TFNP than non-cancer cells. The cytotoxic assay of TFNP against MDA-MB-231 cells and 293T cells was nontoxic to both cell lines compared with Trisindolina-5 which was toxic. Folate receptor targeting by TFNP gave significant results against MDA-MB-231 cells after 48 hours incubation. TFNP was able to induce cell death at the lowest concentration of the cytotoxic assay performed.
