Abstract

Mikroalga merupakan kelompok mikroorganisme autotrofik yang terbagi menjadi kelompok prokariotik (seperti cyanobacteria) dan eukariotik (seperti diatom, dinoflagellata, dan alga hijau). Kemampuan mikroalga untuk tumbuh dalam kondisi lingkungan yang beragam memungkinkan identifikasi spesies mikroalga dengan profil asam lemak yang bervariasi. Belum adanya kultur koleksi isolat mikroalga dari ITS menjadi salah satu alasan dipilihnya Kolam Asrama ITS sebagai lokasi penelitian. Oleh karena itu, penelitian ini dilakukan dengan tujuan untuk mengidentifikasi spesies mikroalga yang ditemukan dari perairan Kolam Asrama ITS Surabaya menggunakan metode pengamatan morfologi dan identifikasi molekuler serta menguji profil asam lemak dari isolat mikroalga tersebut. Metode yang dilakukan dalam penelitian ini meliputi: (1) identifikasi morfologi mikroalga berdasarkan buku identifikasi, (2) identifikasi molekuler menggunakan marka 16S dan 18S rRNA, serta (3) analisis profil asam lemak mikroalga (%w/w) menggunakan GC-MS. Hasil penelitian menunjukkan bahwa beberapa spesies mikroalga berhasil diidentifikasi dari perairan Kolam Asrama ITS Surabaya, antara lain Dictyosphaerium ehrenbergianum, Vitreochlamys incisa, Desmodesmus armatus, dan Chlorella vulgaris. Profil asam lemak isolat mikroalga bervariasi antar spesies. Dictyosphaerium ehrenbergianum memiliki kadar SFA sebesar 17,57%, MUFA sebesar 12,28%, dan PUFA sebesar 0,33%. Vitreochlamys incisa memiliki kadar SFA sebesar 10,83% dan MUFA sebesar 10,19%. Desmodesmus armatus memiliki kadar SFA sebesar 14,19%, MUFA sebesar 7,39%, dan PUFA sebesar 4,14%. Chlorella vulgaris memiliki kadar SFA sebesar 1,92% dan PUFA sebesar 2,67%. ==============================================================================================================================
Microalgae are a group of autotrophic microorganisms which are divided into prokaryotic groups (such as cyanobacteria) and eukaryotic groups (such as diatoms, dinoflagellates and green algae). The ability of microalgae to grow in diverse environmental conditions allows the identification of microalgae species with varying fatty acid profiles. The absence of a culture collection of microalgae isolates from ITS was one of the reasons for choosing the ITS Dormitory Pond as a research location. Therefore, this research was carried out with the aim of identifying microalgae species found from the waters of the ITS Surabaya Dormitory Pond using morphological observation and molecular identification methods as well as testing the fatty acid profile of the microalgae isolates. The methods used in this research include: (1) morphological identification of microalgae based on the identification book, (2) molecular identification using 16S and 18S rRNA markers, and (3) analysis of the fatty acid profile of microalgae (%w/w) using GC-MS. The research results showed that several species of microalgae were identified from the waters of the ITS Surabaya Dormitory Pond, including Dictyosphaerium ehrenbergianum, Vitreochlamys incisa, Desmodesmus armatus, and Chlorella vulgaris. The fatty acid profile of microalgae isolates varies between species. Dictyosphaerium ehrenbergianum has SFA levels of 17.57%, MUFA of 12.28%, and PUFA of 0.33%. Vitreochlamys incisa has SFA levels of 10.83% and MUFA of 10.19%. Desmodesmus armatus has SFA levels of 14.19%, MUFA of 7.39%, and PUFA of 4.14%. Chlorella vulgaris has SFA levels of 1.92% and PUFA of 2.67%.
