Abstract

Proses restrukturisasi kulit dalam penyembuhan luka dapat diamati secara fisioanatomi melalui kajian profil darah dan histologi kulit salah satunya penilaian kolagenisasi (persentase, pola persebaran dan struktur kolagen). Studi pendahuluan secara in vivo diketahui bahwa pemberian phenylethylamine (PEA) berpotensi mempercepat proses penyembuhan luka sehingga penelitian lanjutan dilakukan untuk mengetahui pengaruh PEA pada proses penyembuhan luka kulit pada Mus musculus (mencit) berdasarkan analisis anatomi-histologi kulit dan darah. Metode penelitian yang digunakan dengan pembuatan luka eksisi menggunakan skin biopsy punch pada mencit yang diteteskan variasi konsentrasi PEA 0 μg/mL(D0); 12,5μg/mL(D1); 25μg/mL(D2); 50μg/mL(D3) setiap hari selama 14 hari dan diamati morfologi luka tiap 2 hari sekali dengan Imitomeasure. Pembuatan preparat apusan darah dengan pewarnaan Giemsa dan preparat histologi menggunakan metode parafin dan pewarnaan Masson’s Trichrome (MT). Hasil penelitian pemberian PEA pada proses penyembuhan luka secara kualitatif dan kuantitatif berdasarkan pengamatan morfologi dan ukuran luas luka, profil dan persentase serat kolagen jaringan kulit secara histologis, profil dan persentase diferensial leukosit, serta analisis semi-kuantitatif profil serat kolagen yang dianalisis secara statistik dengan ANOVA one-way dan Tukey menunjukkan bahwa perlakuan PEA (D1, D2, dan D3) memiliki tingkat penyembuhan luka yang lebih tinggi dilihat pada kematangan serat kolagen skor 12,3 s/d 12,7 daripada tanpa perlakuan PEA (D0=10,7) dan tidak beda signifikan dengan kontrol (K=14). Antar variasi konsentrasi PEA tidak beda signifikan menunjukkan pengaruh yang sama terhadap proses penyembuhan luka hingga sama atau mendekati normal.

The process of skin restructuring in wound healing can be observed physioanatomically through studying blood profiles and skin histology, one of which is assessing collagenization (percentage, distribution pattern and structure of collagen). Preliminary in vivo studies show that administration of phenylethylamine (PEA) has the potential to accelerate the wound healing process, that further research was carried out to determine the effect of PEA on the skin wound healing process in Mus musculus (mice) based on anatomical-histological analysis of skin and blood. The research method used was the creation of excisional wounds using a skin biopsy punch on mice which were instilled with varying concentrations of PEA 0 μg/mL(D0); 12.5μg/mL(D1); 25μg/mL(D2); 50μg/mL(D3) every day for 14 days and the wound morphology was observed every 2 days with Imitomeasure. Blood smear preparations using Giemsa staining and histology preparations using the paraffin method and Masson's Trichrome (MT) staining. The results of research on the administration of PEA in the wound healing process are qualitative and quantitative based on observations of the morphology and size of the wound area, the profile and percentage of collagen fibers in skin tissue histologically, the profile and percentage of differential leukocytes, as well as semi-quantitative analysis of the collagen fiber profile which was analyzed statistically using ANOVA one-way and Tukey showed that PEA treatment (D1, D2, and D3) had a higher rate of wound healing seen from collagen fiber maturity scores of 12.3 to 12.7 than without PEA treatment (D0=10.7) and not significantly different from controls (K=14). There was no significant difference between the variations in PEA concentration, showing the same effect on the wound healing process until it was the same or close to normal.
