Abstract

Meningkatnya kasus infeksi yang menyebabkan naiknya angka kematian di seluruh dunia serta penggunaan antibiotik yang tidak terkendali untuk menangani infeksi bakteri berpotensi menimbulkan adanya resistensi bakteri terhadap antibiotik. Oleh sebab itu, peneliti gencar dalam menggali potensi tumbuhan sebagai antibakteri terbarukan. Pada penelitian ini, bunga cengkeh diuji aktivitas antibakteri terhadap bakteri E. coli dan S. aureus. Cengkeh diekstrak dengan metode maserasi menggunakan empat variasi pelarut (etil asetat, aseton, etanol, dan akuades). Ekstrak yang didapat kemudian diuji kandungan fitokimianya dan antibakteri terhadap bakteri E. coli dan S. aureus secara in vitro dengan metode difusi cakram (kualitatif) dan mikrodilusi (kuantitatif). Rendemen ekstrak paling banyak diperoleh menggunakan pelarut etanol (24%) dan rendemen ekstrak paling sedikit diperoleh menggunakan pelarut akuades (11%). Dari uji kandungan fitokimia diketahui bahwa semua ekstrak mengandung flavonoid, alkaloid, dan tanin tapi tidak mengandung steroid dan terpenoid; saponin dan glikosida hanya terdapat pada ekstrak aseton, etil asetat dan etanol. Melalui uji antibakteri difusi cakram, diketahui bahwa ekstrak etil asetat bunga cengkeh bekerja paling baik dalam menghambat pertumbuhan bakteri E. coli dan S. aureus. Nilai konsentrasi hambat minimum (KHM) ditentukan melalui metode mikrodilusi. Untuk bakteri E. coli, nilai KHM tercapai pada konsentrasi 41,704 mg/mL. Sedangkan untuk bakteri S. aureus, nilai KHM tercapai pada konsentrasi 85,6845 mg/mL

The increasing cases of infections that cause increased mortality worldwide and the uncontrolled use of antibiotics to treat bacterial infections have the potential to cause bacterial resistance to antibiotics. Therefore, researchers are aggressively exploring the potential of plants as renewable antibacterials. In this study, clove flowers were tested for antibacterial activity against E. coli and S. aureus bacteria. Cloves were extracted by maceration method using four solvent variations (ethyl acetate, acetone, ethanol, and distilled water). The extracts obtained were then tested for phytochemical content and antibacterial against E. coli and S. aureus bacteria in vitro by disc diffusion (qualitative) and microdilution (quantitative) methods. The highest extract yield was obtained using ethanol solvent (24%) and the least extract yield was obtained using distilled water solvent (11%). Phytochemical tests showed that all extracts contained flavonoids, alkaloids and tannins but no steroids and terpenoids; saponins and glycosides were only found in acetone, ethyl acetate and ethanol extracts. Through the disc diffusion antibacterial test, it was found that the ethyl acetate extract of clove flower worked best in inhibiting the growth of E. coli and S. aureus bacteria. The minimum inhibitory concentration (KHM) value was determined through the microdilution method. For E. coli bacteria, the KHM value was reached at a concentration of 41.704 mg/mL. While for S. aureus bacteria, the KHM value was reached at a concentration of 85.6845 mg/mL
