Abstract
Minyak atsiri jahe memiliki daya komersil yang cukup tinggi karena banyak dimanfaatkan dalam bidang terapi, aromaterapi, serta industri penyedap makanan. Minyak atsiri jahe dibentuk oleh senyawa volatil yang terkandung di dalam jahe. Pada penelitian ini, akan diekstraksi minyak atsiri dari sampel jahe merah dengan menggunakan metode Microwave Assisted Hydrodistillation (MAHD) yang dapat meminimalkan waktu ekstraksi serta pelarut melalui pendekatan Response Surface Methodology (RSM) untuk melihat keterkaitan dan pengaruh dari masing-masing variabel. Penelitian ini bertujuan untuk mengetahui pengaruh variasi daya microwave, solvent to feed ratio (SFR), dan waktu ekstraksi terhadap nilai yield, kandungan senyawa bioaktif zingiberene, dan total phenolic content serta aktivitas antioksidan dari minyak atsiri jahe. Metode penelitian yang digunakan adalah desain eksperimen menggunakan RSM dengan model Box-Behnken Design (BBD) dengan melakukan variasi pada daya microwave, solvent to feed ratio (SFR), dan waktu ekstraksi. Hasil penelitian menunjukkan bahwa nilai yield minyak atsiri jahe yang diperoleh berada pada rentang 0,72 hingga 2,62%, nilai yield tertinggi diperoleh pada kondisi daya microwave 450 Watt, SFR 1:12, dan waktu ekstraksi 120 menit sebesar 2,62%. Nilai kadar zingiberene minyak atsiri jahe yang diperoleh berada pada rentang 21,34 hingga 31,17% dengan nilai kadar zingiberene tertinggi terdapat pada kondisi daya microwave 450 Watt, SFR 1:12, dan waktu ekstraksi 120 menit sebesar 31,17%. Nilai TPC minyak atsiri jahe berada pada rentang 2102,74 ± 25,420 hingga 3367,00 ± 21,03 mg GAE/100 g minyak atsiri, nilai TPC tertinggi terdapat pada minyak atsiri jahe sampel P2S3T3 dengan kondisi daya microwave 450 Watt, SFR 1:12, dan waktu ekstraksi 120 menit yaitu sebesar 3369,02 ± 21,027 mg GAE/ 100 g minyak atsiri. Nilai IC50 pada minyak atsiri jahe berada pada rentang 130,185 hingga 150,160 ppm, nilai IC50 tertinggi terdapat pada minyak atsiri jahe sampel P2S3T3 dengan kondisi daya microwave 450 Watt, SFR 1:12, dan waktu ekstraksi selama 120 menit sebesar 130,185 ppm. Minyak atsiri jahe pada sampel P2S3T3 memiliki nilai yield, kandungan zingiberene, nilai TPC, dan aktivitas antioksidan IC50 yang paling baik. Berdasarkan hasil yang diperoleh daya microwave menjadi variabel yang berpengaruh secara signifikan terhadap yield, kandungan senyawa bioaktif zingiberene, dan total phenolic content serta aktivitas antioksidan dari minyak atsiri jahe.

Ginger essential oil has a high commercial value because it is widely used in therapy, aromatherapy, and food flavoring industries. Ginger essential oil is formed by volatile compounds contained in ginger. In this study, essential oil will be extracted from red ginger samples using Microwave Assisted Hydrodistillation (MAHD) method that can minimize extraction time and solvent through Response Surface Methodology (RSM) approach to see the relationship and influence of each variable. This study aims to determine the effect of variations in microwave power, solvent to feed ratio (SFR), and extraction time on yield value, content of bioactive zingiberene compounds, and total phenolic content and antioxidant activity of ginger essential oil. The research method used is experimental design using RSM with Box-Behnken Design (BBD) model by varying microwave power, solvent to feed ratio (SFR), and extraction time. The results showed that the yield value of ginger essential oil obtained was in the range of 0.72 to 2.62%, the highest yield value was obtained in the conditions of 450 Watt microwave power, SFR 1:12, and 120 minutes extraction time of 2.62%. The zingiberene content value of ginger essential oil obtained was in the range of 21.34 to 31.17% with the highest zingiberene content value found in the conditions of 450 Watt microwave power, SFR 1:12, and 120 minutes extraction time of 31.17%. The TPC value of ginger essential oil is in the range of 2102.74 ± 25.420 to 3367.00 ± 21.03 mg GAE/100 g essential oil, the highest TPC value is found in ginger essential oil sample P2S3T3 with microwave power conditions of 450 Watts, SFR 1:12, and extraction time of 120 minutes which is 3369.02 ± 21.027 mg GAE / 100 g essential oil. The IC50 value of ginger essential oil is in the range of 130.185 to 150.160 ppm, the highest IC50 value is found in ginger essential oil sample P2S3T3 with microwave power conditions of 450 Watt, SFR 1:12, and extraction time for 120 minutes at 130.185 ppm. Ginger essential oil in sample P2S3T3 has the best yield value, zingiberene content, TPC value, and IC50 antioxidant activity. Based on the results obtained, microwave power is a variable that significantly affects the yield, content of bioactive compounds zingiberene, and total phenolic content and antioxidant activity of ginger essential oil.
