Abstract

Desmodesmus sp. merupakan salah satu mikroalga yang mempunyai berbagai potensi, salah satunya sebagai biofuel. Keberadaan koleksi kultur menggunakan kriopreservasi menjadi metode yang banyak digunakan dalam menjaga ketersediaan mikroorganisme dengan segala potensinya. Penggunaan freezer lemari pendingin berpotensi sebagai metode preservasi beku yang sederhana dengan penggunaan konsentrasi senyawa protektan yang tepat untuk menjaga viabilitas sel. Pada penelitian ini dipelajari pengaruh konsentrasi senyawa protektan dimetil sulfoksida (DMSO) dan durasi preservasi beku terhadap viabilitas sel Desmodesmus sp. Konsentrasi DMSO yang digunakan adalah 5%, 8%, dan 10% dengan durasi preservasi meliputi 7, 14, 21, dan 28 hari. Parameter viabilitas yang diamati meliputi grafik pertumbuhan, laju pertumbuhan, dan waktu generasi. Data yang diperoleh dianalisis secara deskriptif kuantitatif dan secara statistik. Hasil penelitian menunjukkan bahwa variasi durasi preservasi dan konsentrasi DMSO berpengaruh terhadap grafik pertumbuhan yaitu preservasi 7 hari menunjukkan rata-rata fase eksponensial lebih cepat dibandingkan rata-rata eksponensial durasi preservasi 14, 21, dan 28 hari. Variasi durasi preservasi dan konsentrasi DMSO berpengaruh terhadap laju pertumbuhan yaitu 5% DMSO dan durasi preservasi 28 hari menunjukkan laju pertumbuhan tertinggi dibandingkan variasi 8% dan 10% DMSO serta durasi preservasi 7, 14, dan 21 hari. Variasi durasi preservasi dan konsentrasi DMSO berpengaruh terhadap waktu generasi yaitu 5% DMSO dan durasi preservasi 14 hari menunjukkan waktu generasi tercepat dibandingkan variasi 8% dan 10% DMSO serta durasi preservasi 7, 21, dan 28 hari.

Desmodesmus sp. is a microalga that has various potentials, one of which is as biofuel. The existence of culture collections using frozen preservation is a method that is widely used to maintain the availability of microorganisms with all their potential. The use of refrigerator freezers has the potential to be a simple frozen preservation method by using appropriate concentrations of protectant compounds to maintain cell viability. In this research, we studied the effect of the concentration of the protectant compound dimethyl sulfoxide (DMSO) and the duration of frozen preservation on the cell viability of Desmodesmus sp. The DMSO concentrations used were 5%, 8%, and 10% with preservation durations including 7, 14, 21, and 28 days. The viability parameters observed include growth graphs, growth rates, and generation times. The data obtained was analyzed descriptively, quantitatively and statistically. The results showed that variations in preservation duration and DMSO concentration influenced the growth graph, namely that 7 days of preservation showed an average exponential phase that was faster than the exponential average of preservation durations of 14, 21 and 28 days. Variations in preservation duration and DMSO concentration influenced growth rate, namely 5% DMSO and a preservation duration of 28 days showed the highest growth rate compared to variations of 8% and 10% DMSO and preservation durations of 7, 14 and 21 days. Variations in preservation duration and DMSO concentration influenced generation time, namely 5% DMSO and a preservation duration of 14 days showed the fastest generation time compared to variations of 8% and 10% DMSO and preservation durations of 7, 21 and 28 days.
