Abstract

Polyunsaturated Fatty Acid (PUFA) adalah asam lemak dengan jumlah ikatan rangkap
dua (double bond) sebanyak lebih dari satu. Manfaat PUFA sangat krusial bagi tubuh yaitu
menjaga fluiditas dari dinding sel dan mengatasi berbagai penyakit seperti penyakit
kardiovaskular, kanker, demesia, dan alzheimer. PUFA tidak dapat diproduksi secara de novo
di dalam tubuh sehingga perlu asupan dari makanan, seperti ikan laut. Selain ikan, PUFA
juga di produksi de novo oleh bakteri melalui polyketide synthase enzyme (PKS) yang
melibatkan enzim desaturase yang dideteksi keberadaannya dengan Triphenyl Tetrazolium
Chloride (TTC). Penelitian ini bertujuan untuk mengisolasi bakteri pereduksi Tryiphenyl
tetrazolium Chloride (TTC) di kawasan Mangrove Wonorejo sebagai penanda awal penghasil
PUFA serta mengidentifikasi genus dari bakteri tersebut. Sampling dilakukan dengan metode
line transect dengan sampel dikompositkan. Isolasi dilakukan dengan metode pour plate
dengan media Nutrient Agar (NA). Purifikasi dilakukan dengan metode streak plate 16
goresan pada media NA. Identifikasi dilakukan secara konvensional yaitu dengan menguji
karakter fenetik seperti morfologi, fisiologi, dan biokimia. Analisis hasil akan dibuat dalam
bentuk Generic assignment dengan mencocokkan sifat-sifat isolat bakteri yang didapat
berdasarkan acuan Bergey’s Manual of Determinative Bacteriology edisi ke-8. Genus bakteri
yang didapat adalah Neisseria, Acinetobacter, diduga Brucella, Pseudomonas, dan Moraxella.

Polyunsaturated Fatty Acid (PUFA) is a fatty acid with more than one double bond. The benefits of PUFA are crucial for the body, namely maintaining the fluidity of cell walls and overcoming various diseases such as cardiovascular disease, cancer, dementia, and Alzheimer's. PUFA cannot be produced de novo in the body so it needs intake from food, such as sea fish. In addition to fish, PUFA is also produced de novo by bacteria through the polyketide synthase enzyme (PKS) which involves the desaturase enzyme whose presence is detected with Triphenyl Tetrazolium Chloride (TTC). This study aims to isolate Tryiphenyl Tetrazolium Chloride (TTC) reducing bacteria in the Wonorejo Mangrove area as an early marker of PUFA producers and to identify the genus of these bacteria. Sampling was carried out using the line transect method with composite samples. Isolation was carried out using the pour plate method with Nutrient Agar (NA) media. Purification was carried out using the streak plate method with 16 scratches on NA media. Identification was carried out conventionally by testing phenetic characters such as morphology, physiology, and biochemistry. Analysis of the results will be made in the form of Generic assignment by matching the properties of the bacterial isolates obtained based on the Bergey's Manual of Determinative Bacteriology 8th edition reference. The bacterial genera obtained were Neisseria, Acinetobacter, suspected Brucella, Pseudomonas, and Moraxella.
