Abstract

Penelitian senyawa antibakteri menjadi salah satu penelitian yang masih banyak diminati. Hal ini terjadi karena kecenderungan resistensi suatu bakteri terhadap obat, sehingga diperlukan pengembangan obat baru dengan harapan dapat memberikan suatu obat yang sudah diatas standar. Metallodrugs mreupakan salah satu alternatif perancangan obat baru dalam melawan resistensi. Telah berhasil dilakukan sintesis ligan turunan hidrazon berbasis benzohidrazida, yaitu 2-hidroksibenziliden benzohidrazida berupa padatan berwarna putih keruh dengan rendemen sebesar 62,85% Dalam penelitian ini, diperoleh suatu kompleks Zn(II)-2- hidroksibenziliden benzohidrazida yang diperoleh dari hasil reaksi ligan 2-hidroksibenziliden benzohidrazida dengan logam Zn(II) berupa padatan berwana hijau kekuningan dengan rendemen sebesar 60,68%. Perbandingan logam dan ligan yaitu 1:2. Kemudian, ligan dan kompleks di karakterisasi TGA/DTA, CHNS, UV-Vis, FTIR, XRD, NMR, dan MS serta uji antibakteri terhadap bakteri gram positif (S.aureus) dan bakteri gram negatif (E.coli). Didapatkan hasil uji antibakteri ligan dan kompleks berupa rata-rata zona inhibisi. Nilai rata- rata zona inhibisi pada ligan sebesar 7,15 ± 1,05 mm untuk S.aureus dan 8,6 ± 0,5 mm untuk E.coli. Nilai rata-rata zona inhibisi pada kompleks sebesar 12,1 ± 0 mm untuk S.aureus dan 12,05 ± 0,05 mm untuk E.coli. Nilai zona inhibisi pada senyawa kompleks mengalami peningkatan yang lebih baik dibandingkan dengan nilai zona inhibisi ligan.

Research on antibacterial compounds is one of the studies that is still in great demand. This happens because of the tendency of a bacterium to resist drugs, so it is necessary to develop new drugs in the hope of providing a drug that is already above the standard. Metallodrugs are one of the alternatives to designing new drugs to fight resistance. The synthesis of benzohydride-based hydrazone derivative ligands has been successfully carried out, namely 2- hydroxybenzidine benzohydride in the form of a cloudy white solid with a yield of 62,85% In this study, a Zn(II)-2-hydroxybenzidine benzohydride complex was obtained from the reaction of the ligand 2-hydroxybenzidine benzohydride with Zn(II) metal in the form of a yellowish- green solid with a yield of 60,68%. The ratio of metals and ligands is 1:2. Then, ligands and complexes were characterized by TGA/DTA, CHNS, UV-Vis, FTIR, XRD, NMR, and MS as well as antibacterial tests against gram-positive bacteria (S.aureus) and gram-negative bacteria (E.coli). The results of the ligand and complex antibacterial test were obtained in the form of the average inhibition zone. The average value of the inhibition zone in the ligand was 7.15 ±
1.05 mm for S.aureus and 8.6 ± 0.5 mm for E.coli. The average value of the inhibition zone in the complex was 12.1 ± 0 mm for S.aureus and 12.05 ± 0.05 mm for E.coli. The value of the inhibition zone in complex compounds has increased better compared to the value of the ligand inhibition zone.
