Abstract

Limbah organik cair dan padat yang dihasilkan oleh berbagai industri, seperti tekstil, makanan, dan farmasi, mengandung pewarna sintetis berbahaya yang sulit diuraikan melalui proses biologi konvensional. Metilen biru (MB), salah satu pewarna kationik, telah menjadi perhatian karena dampaknya terhadap kesehatan manusia dan tantangan dalam dekontaminasinya. Oleh karena itu diperlukan metode untuk mendegradasi limbah MB. Proses biodegradasi menggunakan mikroorganisme menawarkan solusi ramah lingkungan untuk pengolahan limbah ini. Bakteri Uncultured Bacterium strain BS2049 (JN4), yang diisolasi dari lahar Gunung Semeru, memiliki potensi dalam mendekolorisasi MB. Tujuan penelitian ini adalah mengetahui kemampuan isolat bakteri JN4 yang diimobilisasi ke dalam matriks SA-PVA dalam mendegradasi MB dan mengkarakterisasi beads yang dihasilkan. Dekolorisasi metilen biru (MB) dilakukan menggunakan larutan pewarna dengan konsentrasi 10 mg/L melalui dua metode yaitu free cell dan imobilisasi, baik untuk bakteri hidup maupun mati, selama masa inkubasi 10 hari. Hasil penelitian menunjukkan bahwa metode imobilisasi lebih efektif dibandingkan metode free cell. Persentase dekolorisasi dengan imobilisasi SA-PVA-JN4 dalam kondisi hidup, mati, dan tanpa bakteri masing-masing mencapai 89,704%, 62,054%, dan 52,430%. Sementara itu, metode free cell menghasilkan persentase dekolorisasi yang lebih rendah, yaitu 64,095% untuk bakteri hidup dan 40,648% untuk bakteri mati. Hal ini dapat membuktikan bahwa SA/PVA/JN4 memiliki kemampuan dalam mendekolorisasi dengan persentase yang signifikan dan berfungsi sebagai alternatif untuk mengatasi limbah pewarna MB secara efektif dan efisien.

Liquid and solid organic waste generated by various industries, such as textile, food, and pharmaceutical, contain hazardous synthetic dyes that are difficult to decompose through conventional biological processes. Methylene blue (MB), one of the cationic dyes, has become a concern due to its impact on human health and challenges in its decontamination. Therefore, a method is needed to degrade MB waste. The biodegradation process using microorganisms offers an environmentally friendly solution for the treatment of this waste. Uncultured Bacterium strain BS2049 (JN4), isolated from Mount Semeru lava, has the potential to decolorize MB. The purpose of this study was to determine the ability of JN4 bacterial isolates immobilized into SA-PVA matrix to degrade MB and to characterize the resulting beads. Decolorization of methylene blue (MB) was carried out using a dye solution with a concentration of 10 mg/L through two methods, namely free cell and immobilization, for both live and dead bacteria, during an incubation period of 10 days. The results showed that the immobilization method was more effective than the free cell method. The percentage of decolorization with immobilization of SA-PVA-JN4 in live, dead, and without bacteria conditions reached 89.704%, 62.054%, and 52.430%, respectively. Meanwhile, the free cell method produced a lower percentage of decolorization, which was 64.095% for live bacteria and 40.648% for dead bacteria. This can prove that SA/PVA/JN4 has the ability to decolorize with a significant percentage and functions as an alternative to overcome MB dye waste effectively and efficiently
