Abstract
Keanekaragaman hayati di ekosistem perairan, khususnya bakteri, berperan penting dalam menjaga keseimbangan dan keberlanjutan ekosistem. Keanekaragaman tersebut mencakup fenotip dan genotip yang dipengaruhi oleh faktor gen dan ekologi. Bakteri berperan sebagai penyumbang terbesar dalam ekosistem karena kemampuan adaptasi di berbagai habitat terutama di perairan. Bakteri di habitat air memiliki keanekaragaman hayati karena keadaan lingkungannya yang berbeda. Komunitas bakteri di perairan juga dapat dipengaruhi adanya fraksi logam berat seperti fraksi timbal (Pb). Tujuan dari penelitian ini untuk mengetahui keanekaragaman bakteri resisten timbal (Pb) dari perairan sungai Wonorejo dengan pendekatan taksonomi numerik fenetik. Penelitian dilakukan dengan mengambil sampel air di titik 1 dengan lokasi sungai jembatan tambangan Wonorejo (7°18'28"S 112°47'55"E) dan titik 2 dengan lokasi sungai Kawasan Ekosistem Mangrove Wonorejo (7°18'26"S 112°49'23"E). Penelitian ini dilakukan dengan metode klasifikasi taksonomi numerik fenetik berdasarkan karakter fenotipe bakteri, seperti morfologi koloni dan sel, serta uji biokimia dan fisiologi. Hasil penelitian ini diperoleh 8 isolat yang resisten logam Pb dengan konsentrasi hingga 150 ppm. Pada konstruksi dendogram didapatkan 3 cluster dengan rentang indeks similaritasnya sebesar 0,467 hingga 0,911. Dari 3 cluster tersebut terdapat 3 genus yang teridentifikasi. Pada cluster A terdapat isolat P4a dengan isolat teridentifikasi sebagai anggota dari genus Bacillus. Pada cluster B terdapat isolat P2a, P2b, dan P4b yang teridentifikasi sebagai anggota dari genus Corynebacterium. Pada cluster C isolat T2, T3, T4, dan T7 yang teridentifikasi sebagai anggota dari genus Staphylococcus.

Biodiversity in aquatic ecosystems, especially bacteria, plays an important role in maintaining ecosystem balance and sustainability. The diversity includes phenotypes and genotypes that are influenced by genes and ecological factors. Bacteria act as the largest contributor to the ecosystem due to their adaptability in various habitats, especially in water. Bacteria in aquatic habitats have biodiversity due to their different environmental conditions. Bacterial communities in waters can also be influenced by the presence of heavy metal fractions such as lead (Pb) fractions. The purpose of this study was to determine the diversity of lead (Pb) resistant bacteria from Wonorejo river using a phenetic numerical taxonomy approach. The research was conducted by taking water samples at location 1 with the location of the Wonorejo pond bridge river (7°18'28 “S 112°47'55 ‘E) and location 2 with the location of the Wonorejo Mangrove Ecosystem Area river (7°18'26 ’S 112°49'23 ”E). This research was conducted with the phenetic numerical taxonomic classification method based on bacterial phenotype characters, such as colony and cell morphology, as well as biochemical and physiological tests. The results of this study obtained 8 isolates resistant to Pb metal with concentrations up to 150 ppm. In the dendrogram construction, 3 clusters were obtained with a similarity index range of 0.467 to 0.911. Of the 3 clusters there are 3 genus identified. In cluster A there is isolate P4a with isolates identified as members of the genus Bacillus. In cluster B there are isolates P2a, P2b, and P4b which are identified as members of the genus Corynebacterium. In cluster C, isolates T2, T3, T4, and T7 were identified as members of the genus Staphylococcus.
