Abstract
Kanker merupakan suatu penyakit yang ditandai dengan adanya pertumbuhan tidak normal dari sel jaringan tubuh. Kanker serviks merupakan penyebab kematian keempat akibat kanker pada wanita, yang sebagian besar disebabkan oleh infeksi HPV onkogenik, khususnya HPV16 dan HPV18. Pengobatan konvensional seperti pembedahan, kemoterapi, dan radioterapi sering menimbulkan efek samping yang signifikan, sehingga alternatif pengobatan lebih aman diperlukan. Salah satu alternatif yang menjanjikan adalah penggunaan nanopartikel perak (AgNPs) yang disintesis dari ekstrak daun dewa (Gynura pseudochina). AgNPs memiliki sifat bioaktif yang dapat menghambat proliferasi sel kanker dan menginduksi apoptosis. Hasil karakterisasi AgNPs yang telah disintesis dari G. pseudochina pada penelitian sebelumnya berada pada panjang gelombang 440 nm dengan ukuran 17–35 nm masing-masing menggunakan Spectrophotometer Uv-Vis dan Transmission Electron Microscopy (TEM). Efektivitas AgNPs terhadap sel HeLa diamati melalui uji sitotoksisitas, analisis apoptosis, serta analisis ekspresi gen c-myc dan Cyclin D. Hasil menunjukkan bahwa AgNPs memiliki aktivitas sitotoksik terhadap sel HeLa dengan nilai IC50 sebesar 33 μg/mL, mampu menginduksi apoptosis sebesar 10,2%, serta menurunkan ekspresi gen c-Myc dan Cyclin D secara signifikan. Dengan demikian, AgNPs dari ekstrak daun dewa menunjukkan potensi sebagai agen antikanker alternatif melalui mekanisme penghambatan proliferasi dan induksi apoptosis pada sel kanker serviks (HeLa).

Cancer is a disease characterized by abnormal growth of body tissue cells. Cervical cancer is the fourth cause of cancer death in women, most of which are caused by oncogenic HPV infection, especially HPV16 and HPV18. Conventional treatments such as surgery, chemotherapy and radiotherapy often cause significant side effects, so safer treatment alternatives are needed. One promising alternative is the use of silver nanoparticles (AgNPs) synthesized from dewa leaf extract (Gynura pseudochina). AgNPs have bioactive properties that can inhibit cancer cell proliferation and induce apoptosis. The results of the characterization of AgNPs that have been synthesized from G. pseudochina in previous studies were at a wavelength of 440 nm with a size of 17–35 nm using a Uv-Vis Spectrophotometer and Transmission Electron Microscopy (TEM), respectively. The effectiveness of AgNPs against HeLa cells was observed through cytotoxicity tests, apoptosis analysis, as well as c-myc and Cyclin D gene expression analysis. The results showed that AgNPs had cytotoxic activity against HeLa cells with an IC50 value of 33 μg/mL, were able to induce apoptosis by 10.2%, and significantly reduce c-Myc and Cyclin D gene expression. Thus, AgNPs from the extract of the dewa leaf show potential as an alternative anticancer agent through the mechanism of inhibition of proliferation and induction of apoptosis in cervical cancer cells (HeLa).
