Abstract

Ganoderma boninense merupakan jamur patogen penyebab penyakit tanaman. Salah satu tanaman yang menjadi inang utamanya adalah kelapa sawit (Elaeis guineensis Jacq.) yang dapat menimbulkan penyakit Busuk Pangkal Batang (BPB). Penyakit ini menurunkan produksi kelapa sawit sebesar 15 ton dengan kerugian mencapai USD 500 juta per tahun. Sinar ultraviolet-C (UV-C) adalah sinar radiasi spektrum elektromagnetik dengan panjang gelombang 200-280 nm yang bersifat fungistatic atau menghambat pertumbuhan jamur, juga memiliki efek germisidal pada panjang gelombang 253,7 nm yang efektif menghambat pertumbuhan mikroorganisme. Efektivitas sinar UV-C dipengaruhi oleh beberapa faktor antara lain posisi alat, durasi, dan jumlah paparan sinar UV-C. Penelitian ini bertujuan untuk mengetahui pengaruh posisi alat, durasi, dan jumlah paparan sinar UV-C dalam menghambat pertumbuhan jamur G. boninense. Isolat jamur G. boninense usia 3 hari di medium PDA dipapar sinar UV-C dengan posisi alat horizontal dan vertikal selama 10, 20, 30, 40, 50, dan 100 detik. Setelah pemaparan, isolat diinkubasi selama 3 hari dan setiap hari diukur diameter koloninya. Selanjutnya, pada usia 6 hari isolat dipapar kembali untuk paparan dua kali kemudian diinkubasi. Inkubasi dihentikan setelah pertumbuhan salah satu isolat telah memenuhi medium PDA di cawan petri. Parameter yang diamati adalah nilai pertumbuhan jamur, persentase penghambatan pertumbuhan jamur, dan morfologi mikroskopis. Hasil menunjukkan bahwa posisi alat horizontal merupakan posisi paling efektif dalam menghambat pertumbuhan G. boninense dibandingkan posisi vertikal. Durasi 100 detik memberikan penghambatan tertinggi dibandingkan durasi yang lebih pendek. Selain itu, jumlah paparan dua kali lebih efektif dibandingkan paparan satu kali. Perlakuan dengan kombinasi posisi horizontal, durasi 100 detik, dan jumlah paparan dua kali menghasilkan penghambatan pertumbuhan paling tinggi baik secara makroskopis dan kerusakan struktur hifa mikroskopis.

Ganoderma boninense is a pathogenic fungus that causes plant diseases. One of the main hosts is oil palm (Elaeis guineensis Jacq.) which can cause Stem Base Decay Disease (BPB). This disease decreases oil palm production by 15 tons with losses reaching USD 500 million per year. Ultraviolet-C (UV-C) light is a radiation from the electromagnetic spectrum with a wavelength of 200-280 nm that is fungistatic or inhibits fungal growth, and also has a germicidal effect at a wavelength of 253.7 nm that effectively inhibits the growth of microorganisms. The effectiveness of UV-C light is influenced by several factors including the position of the equipment, duration, and amount of UV-C exposure. This study aims to determine the effect of the position of the equipment, duration, and amount of UV-C exposure in inhibiting the growth of G. boninense. Isolate of G. boninense aged 3 days on PDA medium was exposed to UV-C light in both horizontal and vertical positions for 10, 20, 30, 40, 50, and 100 seconds. After exposure, the isolate was incubated for 3 days and the diameter of the colonies was measured daily. Subsequently, on day 6, the isolate was exposed again for two more exposures and then incubated. The incubation was stopped after the growth of one of any isolate had filled the PDA medium in the Petri dish. The observed parameters were fungal growth value, percentage of inhibition of fungal growth, and microscopic morphology. The results show that the horizontal position of the tool is the most effective position in inhibiting the growth of G. boninense compared to the vertical position. A duration of 100 seconds provides the highest inhibition compared to shorter durations. Furthermore, exposure twice is more effective than single exposure. Treatment with a combination of horizontal position, 100 seconds duration, and double exposure results in the highest growth inhibition both macroscopically and in causing damage to the microscopic hyphal structure.
