Abstract

Ganoderma boninense merupakan jamur patogen yang menginfeksi kepala sawit dan menyebabkan penurunan produksi sebesar 50-80%. Upaya pengendalian jamur patogen G.boninense telah dilakukan dengan beberapa cara seperti penggunaan fungisida, pengendalian hayati dengan menggunakan mikroorganisme yang bersifat antagonis, dan pembiakan mikoriza. Namun, belum dapat memberikan efektivitas jangka panjang dalam penanganannya. Upaya pengendalian non-kimia yang bersifat ramah lingkungan menjadi alternatif penting, salah satunya menggunakan gelombang ultrasonik. Gelombang ultrasonik merupakan gelombang bunyi dengan frekuensi diatas 20.000 Hz. Penelitian ini bertujuan untuk mengkaji pengaruh frekuensi, waktu, dan jumlah paparan gelombang ultrasonik terhadap pertumbuhan jamur G.boninense secara in vitro. Metode yang dilakukan meliputi pemaparan gelombang ultrasonik pada jamur G.boninense yang tumbuh di dalam cawan petri pada media PDA dengan variasi frekuensi (40 kHz dan 1 MHz), waktu paparan (1, 3, 5 menit), serta jumlah paparan (1 kali dan 2 kali). Parameter yang diamati yaitu diameter pertumbuhan jamur G.boninense dari pertumbuhan radial miselium dan perubahan morfologi hifa menggunakan uji Scanning electron microscope (SEM). Hasil penelitian menunjukkan bahwa gelombang ultrasonik dengan frekuensi 40 kHz dan 1 MHz dapat menghambat pertumbuhan jamur Ganoderma boninense yang juga didukung oleh waktu dan jumlah paparan. Sampel yang memiliki diameter paling kecil yaitu yang diberi paparan gelombang ultrasonik dengan frekuensi 1 MHz selama 5 menit. Kemudian, hasil pengamatan SEM menunjukkan adanya kerusakan struktural pada hifa berupa patahan, gagal mengalami percabangan dan pengerutan.

Ganoderma boninense is a pathogenic fungus that infects oil palm and causes a production decline of 50-80%. Efforts to control the pathogenic fungus G.boninense have been carried out using several methods, such as the use of fungicides, biological control using antagonistic microorganisms, and mycorrhizal breeding. However, these have not been able to provide long-term effectiveness in handling it. Non-chemical control efforts that are environmentally friendly are an important alternative, one of which is using ultrasonic waves. Ultrasonic waves are sound waves with frequencies above 20,000 Hz. This study aims to examine the effect of frequency, time, and amount of ultrasonic wave exposures on the growth of G. boninense fungi in vitro. The method used includes ultrasonic wave exposure to G.boninense fungi growing in petri dishes on PDA media with variations in frequency (40 kHz and 1 MHz), time (1, 3, 5 minutes), and number of exposures (1 time and 2 times). The parameters observed were the growth diameter of G. boninense fungi from radial mycelium and in hyphal morphology using a Scanning Electron Microscope (SEM) test. The results of the study showed that ultrasonic waves with frequencies of 40 kHz and 1 MHz could inhibit the growth of G.boninense, which was also supported by the time and amount of ultrasonic waves exposure. The sample with the smallest diameter was exposed to ultrasonic waves with a frequency of 1 MHz for 5 mniutes. SEM observations showed structural damage to the hyphae in the form of fractures, failure to branch and shrinkage.
