Abstract

Ganoderma boninense merupakan jamur patogen penyebab Busuk Pangkal Batang (BPB) pada kelapa sawit. Infeksi G. boninense tidak hanya memengaruhi tanaman, tetapi juga mengganggu komunitas mikroba tanah dengan menurunkan keanekaragaman bakteri dan jamur. Mikroorganisme tanah memiliki peran penting dalam menjaga kesuburan tanah serta menekan pertumbuhan patogen. Penelitian ini bertujuan untuk menganalisis dampak infeksi G. boninense terhadap keanekaragaman bakteri pada media tanam kelapa sawit menggunakan dua metode, yaitu karakterisasi menurut Bergey’s Manual of Determinative Bacteriology dan analisis Next Generation Sequencing (NGS) berbasis gen 16S rRNA. Sampel tanah terdiri dari tanah steril dan non-steril dan diinkubasi selama 0 dan 2 bulan. Hasil identifikasi melalui karakterisasi menurut Bergey’s Manual of Determinative Bacteriology menunjukkan bahwa pada bulan ke-0 genus yang terdeteksi meliputi Moraxella, Proteus, Neisseria, Vibrio, Alcaligenes, Acinetobacter, Enterococcus, Corynebacterium, dan Bacillus, dengan dominasi filum Proteobacteria. Pada bulan ke-2, genus yang teridentifikasi meliputi Corynebacterium, Bacillus, Staphylococcus, Streptococcus, Serratia, Aeromonas, dan Pseudomonas, dengan dominansi filum Firmicutes. Pada metode NGS, ditemukan 320 genus dengan dominansi filum Actinomycetota pada bulan ke-0, termasuk Bacillus, Paenibacillus, Streptomyces, Pseudarthrobacter, dan Micromonospora. Sementara itu, pada bulan ke-2, di dominansi oleh filum Proteobacteria dengan genus utama seperti Pseudomonas, MND1, Pseudolabrys, Micromonospora, dan Streptomyces. Terdapat dua genus yang terdeteksi secara konsisten pada kedua metode, yaitu Bacillus dan Pseudomonas, di mana kedua genus tersebut memiliki peran penting selama infeksi G. boninense.

Ganoderma boninense is a pathogenic fungus that causes basal stem rot (BPB) in oil palms. G. boninense infection not only affects plants but also disrupts the soil microbial community by reducing the diversity of bacteria and fungi. Soil microorganisms have an important role in maintaining soil fertility and suppressing pathogen growth. This study aims to analyze the impact of G. boninense infection on bacterial diversity in oil palm growing media using two methods, namely characterization according to Bergey's Manual of Determinative Bacteriology and Next Generation Sequencing (NGS) analysis based on the 16S rRNA gene. Soil samples consisted of sterile and non-sterile soil and were incubated for 0 and 2 months. The results of identification through characterization according to Bergey's Manual of Determinative Bacteriology showed that in the 0th month the detected genera included Moraxella, Proteus, Neisseria, Vibrio, Alcaligenes, Acinetobacter, Enterococcus, Corynebacterium, and Bacillus, with the dominance of the Proteobacteria phylum. In the 2nd month, the identified genera included Corynebacterium, Bacillus, Staphylococcus, Streptococcus, Serratia, Aeromonas, and Pseudomonas, with the dominance of the Firmicutes phylum. In the NGS method, 320 genera were found with the dominance of the Actinomycetota phylum in the 0th month, including Bacillus, Paenibacillus, Streptomyces, Pseudarthrobacter, and Micromonospora. Meanwhile, in the 2nd month, the dominance was by the Proteobacteria phylum with the main genera such as Pseudomonas, MND1, Pseudolabrys, Micromonospora, and Streptomyces. There were two genera that were consistently detected in both methods, namely Bacillus and Pseudomonas, where both genera have important roles during
G. boninense infection.
