Abstract

Terdapat 9.600 jenis tanaman obat di Indonesia, salah satunya adalah keji beling (Strobilanthes crispus). Pada tanaman ini terdapat senyawa kimia seperti flavonoid, alkaloid, terpenoid, saponin, mineral dan vitamin yang bermanfaat bagi manusia. Kultur in vitro merupakan teknik perbanyakan tanaman dengan keunggulan dapat menghasilkan tanaman unggul yang cepat disertai peningkatan kandungan metabolit sekunder menggunakan elisitor asam salisilat (AS). AS merupakan elisitor abiotik yang mampu memodulasi respon fisiologis tanaman. Tujuan penelitian ini adalah mengetahui pengaruh pemberian asam salisilat terhadap pertumbuhan dan profil metabolit Strobilanthes crispus secara in vitro. Penelitian ini menggunakan eksplan berupa nodus aksenik S. crispus yang selanjutnya dikulturkan pada media dasar Murashige and Skoog (MS) disertai penambahan hormon BAP 3 ppm serta NAA 1 ppm dan variasi konsentrasi AS yang berbeda (0, 0.5, 1, 1.5 µM) selama 60 hari. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan 6 pengulangan. Data respon organogenesis, kalogenesis dan profil metabolit dianalisis secara deskriptif kualitatif. Sedangkan data respon pertumbuhan lainnya (jumlah akar, daun, nodus dan tinggi tanaman) dianalisa menggunakan One Way ANOVA. Hasil penelitian menunjukkan bahwa AS mampu menginduksi kalogenesis dan terbentuknya tunas pada eksplan nodus aksenik S. crispus. Perlakuan AS pada konsentrasi (0.5, 1, 1.5 µM) berdampak pada penghambatan di semua parameter pertumbuhan, kecuali diameter kalus dan berat basah. Selain itu, pemberian AS menghasilkan variasi profil metabolit, dengan variasi terbanyak terdapat pada konsentrasi 1 µM dan peak area tertingginya 3,6318% untuk senyawa lupeol (terpenoid).

There are 9,600 types of medicinal plants in Indonesia, one of which is keji beling (Strobilanthes crispus). This plant contains chemical compounds such as flavonoids, alkaloids, terpenoids, saponins, minerals, and vitamins that are beneficial to humans. In vitro culture is a plant propagation technique with the advantage of rapidly producing superior plants and increasing secondary metabolite content using salicylic acid (SA) as an elicitor. SA is an abiotic elicitor capable of modulating plant physiological responses. This study aimed to determine the effect of SA administration on growth and metabolite profiles. This study used axenic nodes of S.crispus as explants. S. crispus which were then cultured on Murashige and Skoog (MS) base media with the addition of 3 ppm BAP hormone and 1 ppm NAA and different AS concentration variations (0, 0.5, 1, 1.5 µM) for 60 days. This study used a completely randomized design (CRD) with 6 replications. Data on organogenesis, callogenesis, and metabolite profiles were analyzed descriptively and qualitatively. Meanwhile, other growth response data (number of roots, leaves, nodes, and plant height) were analyzed using one-way ANOVA. The results of the study showed that SA was able to induce callogenesis and shoot formation in axenic node explants of S. crispus. SA treatment at the given concentration range also had an impact on inhibition in all growth parameters, except callus diameter and fresh weight. Furthermore, AS administration also had an impact on the variation of the resulting metabolite profile, with the greatest variation occurring at a concentration of 1 µM and a peak area of 3.6318% for terpenoid compounds (lupeol).
