Abstract
Beras merupakan makanan pokok utama di Indonesia dan dikenal mengandung senyawa bioaktif, terutama pada varietas beras berpigmen. Senyawa aktif seperti fenolat, flavonoid, dan antosianin memiliki potensi sebagai antioksidan yang penting dalam menangkal radikal bebas dan mencegah penyakit degeneratif. Penelitian ini bertujuan untuk mengevaluasi pengaruh variasi konsentrasi pelarut metanol terhadap kandungan total fenolat, flavonoid, dan aktivitas antioksidan ekstrak biji beras menggunakan metode maserasi. Sampel biji beras A, C, D, J, dan P diekstraksi dengan pelarut metanol pada konsentrasi 20, 40, 60, dan 80 %, kemudian dianalisis kandungan senyawa fenolat menggunakan metode Folin-Ciocalteu dan kandungan flavonoid menggunakan metode pengompleks Al serta aktivitas antioksidan dengan metode DPPH dan ABTS. Hasil menunjukkan bahwa sampel J memiliki kandungan senyawa bioaktif dan aktivitas antioksidan tertinggi, dengan nilai TPC sebesar 50,02 µg AGE/g, TFC sebesar 40,57 µg KE/g, serta aktivitas antioksidan sebesar 71,93% (DPPH) dan 42,62% (ABTS). Diikuti oleh sampel P, dan D yang juga menunjukkan aktivitas tinggi, sementara sampel C memiliki kandungan senyawa bioaktif dan aktivitas antioksidan terendah. Korelasi positif ditemukan antara tingginya kadar TPC dan TFC terhadap aktivitas antioksidan, namun hubungan ini tidak selalu linier pada seluruh sampel. Hal ini dapat disebabkan oleh degradasi senyawa aktif atau perbedaan komposisi senyawa non-fenolat. Hasil ini memperkuat potensi beras berwarna sebagai sumber antioksidan alami serta pentingnya mempertimbangkan jenis varietas dalam proses ekstraksi senyawa bioaktif.

Rice is the main staple food in Indonesia and is known to contain bioactive compounds, especially in pigmented rice varieties. Active compounds such as phenols, flavonoids, and anthocyanins have potential as important antioxidants in counteracting free radicals and preventing degenerative diseases. This study aims to evaluate the effect of varying methanol solvent concentrations on the total phenolic content, flavonoids, and antioxidant activity of rice seed extracts using the maceration method. Rice seed samples A, C, D, J, and P were extracted with methanol solvent at concentrations of 20, 40, 60, and 80 %, then analyzed for phenolic compound content using the Folin-Ciocalteu method, flavonoid content using the Al complexation method, and antioxidant activity using the DPPH and ABTS methods. The results showed that sample J had the highest bioactive compound content and antioxidant activity, with TPC values is 50,02 µg GAE/g, TFC is 40,57 µg QE/g, and antioxidant activity of 71,93% (DPPH) and 42,62% (ABTS). This was followed by samples P and D, which also showed high activity, while sample C had the lowest bioactive compound content and antioxidant capacity. A positive correlation was found between high TPC and TFC levels and antioxidant capacity; however, this relationship was not always linear across all samples. This may be due to the degradation of active compounds or differences in the composition of non-phenolic compounds. These results reinforce the potential of colored rice as a natural antioxidant source and the importance of considering variety type in the extraction process of bioactive compounds.
