Abstract
Berawal dari peristiwa viral produk mengandung babi pada 1988 dan 2000–2009, berbagai metode dikembangkan untuk mendeteksi keaslian produk halal. Salah satunya adalah teknologi ekstraksi DNA menggunakan metode ultrasound-assisted extraction (UAE) yang lebih cepat, hemat pelarut, dan meningkatkan kualitas ekstrak. Penelitian ini mengusulkan inovasi ekstraksi DNA babi dengan UAE menggunakan desain eksperimen full factorial dengan daya 9 dan 450 Watt serta waktu 10 dan 40 menit. Sampel yang digunakan meliputi sosis babi (Cap Sulawesi), kosmetik (Elizavecca Pig Collagen Green Cream), dan obat (Detusitrol). Proses lisis menggunakan ultrasound, sedangkan separasi DNA memakai fenol-kloroform-isoamil alkohol (25:24:1), dan presipitasi DNA menggunakan ammonium asetat 10 M, isopropanol dingin, etanol 70%, serta larutan TE. Hasil menunjukkan perlakuan dengan daya rendah (9 Watt) dan waktu singkat (10 menit) memberikan hasil ekstraksi DNA terbaik, ditunjukkan oleh konsentrasi DNA tinggi dan nilai Cq rendah. Dengan demikian, kombinasi ini efektif menjaga integritas DNA target tanpa degradasi berlebih, mendukung deteksi DNA babi secara lebih akurat dan efisien pada produk pangan, obat, dan kosmetik. Hasil penelitian menunjukkan perlakuan dengan daya rendah (9 Watt) dan waktu singkat (10 menit) menghasilkan crude DNA tertinggi pada sosis babi (9,29 ± 0,39 ng/μL, persentase 58,03%) dan nilai Cq terendah (26,39 ± 1,38), menandakan keberhasilan ekstraksi DNA babi secara optimal. Pada sampel obat dan kosmetik, meskipun konsentrasi DNA lebih rendah, persentase ekstraksi sangat tinggi (1196,77% dan 301,14%). Analisis ANOVA menunjukkan tidak berpengaruh signifikan (p < 0,05) terhadap hasil ekstraksi, sedangkan daya dan waktu tidak signifikan secara mandiri.

Triggered by cases of pork contamination in Indonesian products in 1988 and 2000–2009, various methods were developed to verify halal authenticity. One approach is DNA extraction using ultrasound-assisted extraction (UAE), offering faster, solvent-saving, and higher-quality extraction. This study proposes an innovative pork DNA extraction method using UAE with a full factorial design, involving power levels of 9 and 450 Watts and extraction times of 10 and 40 minutes. Samples included pork sausage (Cap Sulawesi), cosmetic (Elizavecca Pig Collagen Green Cream), and medicine (Detusitrol). Lysis was performed with ultrasound, DNA separation was performed using phenol-chloroform-isoamyl alcohol (25:24:1), and precipitation was employed using 10 M ammonium acetate, cold isopropanol, 70% ethanol, and TE buffer. Results showed that low power (9 Watts) and short time (10 minutes) achieved the best DNA extraction performance, indicated by high DNA concentration and low Cq values. This combination effectively preserves DNA integrity without excessive degradation, thereby supporting accurate and efficient detection of pork DNA in food, pharmaceuticals, and cosmetics. The results showed that treatment with low power (9 Watts) and a short time (10 minutes) produced the highest DNA crude in pork sausages (9.29 ± 0.39 ng/μL, 58.03% yield, and the lowest Cq value (26.39 ± 1.38)), indicating optimal success in pig DNA extraction. In drug and cosmetic samples, although the DNA concentration was lower, the extraction percentage was very high (1196.77% and 301.14%). ANOVA analysis showed no significant effect (p < 0.05) on extraction results, while power and time were not significant independently.
