Abstract

Salah satu permasalahan dalam budidaya ikan adalah penyakit bakterial Edwardsiella ictaluri yang menyebabkan penyakit sistemik Enteric Septicaemia of Catfish (ESC). Pengendalian penyakit telah dilakukan dengan berbagai metode (kimia sintetik dan antibiotik). Penggunaan antibiotik dapat menimbulkan dampak negatif seperti residu obat pada daging dan resistensi bakteri. Probiotik dapat digunakan sebagai metode alternatif pengendalian bakterial. Tujuan penelitian ini adalah isolasi dan skrining bakteri asam laktat (BAL) yang memiliki aktivitas antimikroba E. ictaluri. dentifikasi bakteri digunakan metode gen 16s rRNA. Uji simulasi oleransi cairan lambung dilakukan dengan menghitung viabilitas dan prosentase penempelan BAL pada lendir usus ikan lele Dumbo (Clarias gariepinus). Uji resistensi antibiotik digunakan metode Kirby Bauer. Uji patogenitas menggunakan injeksi intraperitoneal. Sampling ikan dilakukan di Sungai Brantas dengan metode cast net diperoleh 9 ekor. Hasil isolasi diperoleh 29 isolat BAL selanjutnya dipilih 6 isolat berdasarkan aktivitas antimikroba. Hasil identifikasi dan tingkat kesamaan adalah Enterococcus faecalis NBRC100481 (tingkat kesamaan 99,89%), Lactococcus lactis NBRC100933 (99,93% kesamaan), Enterococcus hirae DSM20160 (100%), Enterococcus faecalis ATCC19433 (99,93%), Weissella confusa JCM1093 (100%), dan Weissella cibaria II-I-59 (99,79%). Uji simulasi toleransi cairan lambung menunjukkan viabilitas yang sama secara signifikan (P<0,05) kecuali E. hirae. Prosentase penempelan di lendir usus ikan sebesar 3,42-16,72%. Dua BAL resisten terhadap salah satu dari erythromycin, oxytetrasiklin dan novobiocin yaitu E. faecalis ATCC19433 dan W. cibaria II-I-59. Uji patogenitas menunjukkan keenam BAL non patogen terhadap ikan lele. Bakteri L. lactis NBRC100933, E. hirae DSM 20160 dan W. confusa JCM 1093 berpotensi sebagai kandidat probiotik untuk mencegah penyakit ESC.

One of the problems in fish farming is the bacterial disease Edwardsiella ictaluri which causes systemic bacterial disease Enteric Septicemia of Catfish (ESC). Disease control has been carried out by various methods (synthetic
chemicals and antibiotics). Antibiotics application to control disease can cause negative impacts such as drug residues on meat and bacterial resistance. Probiotics can be used as an alternative method to control pathogen. Objective of this study was to isolate and to screen lactic acid bacteria (LAB) which has antimicrobial activity to E. ictaluri. These isolates were identified using 16s rRNA. In vitro tests assessing tolerance of gastric and intestinal fluids were carried out by calculating the viability, also adhesion LAB on intestinal mucus of catfish (Clarias gariepinus). Antibiotic resistance determined using Kirby Bauer method. Pathogenicity was done by intraperitoneal injection. Nine catfish used as sample caught from Brantas river with cast net method. 29 LAB obtained from isolation and 6 isolates were selected based on their antimicrobial activity. Six selecting LAB identified as Enterococcus faecalis NBRC 100481 (similarity 99.89%), Lactococcus lactis NBRC 100933 (99.93% similarity), Enterococcus hirae DSM 20160 (100%), Enterococcus faecalis ATCC 19433 (99.93%), Weissella confusa JCM 1093 (100%), and Weissella cibaria II-I-59 (99.79%). Tolerance of gastric fluid showed non-significant (P<0.05) except for E. hirae. Adhesion in the intestinal mucus of catfish was 3.42-16.72%. E. faecalis ATCC19433 and W. cibaria II-I-59 resistant to novobiocin and enrofloxacin, respectively. Six LAB were determined to be nonpathogenic to catfish. L. lactis NBRC100933, E. hirae DSM 20160 and W. confusa JCM 1093 were selected as promising probiotic candidates to prevent ESC disease.
