Abstract

Senyawa hidrazon memiliki potensi sebagai antikanker. Diperlukan adanya pengujian lebih lanjut mengenai aktivitas senyawa hidrazon berbasis benzahdehida dan benzohidrasida terhadap Cancer Stem Cell (CSC). CSC dapat digunakan sebagai biomarka untuk deteksi dini tumor, menentukan prognosis dan prediksi respons terhadap terapi sedangkan terapi target terhadap CSC dapat dikembangkan menjadi terapi anti kanker yang berpotensi untuk menyembuhkan. Penelitian ini bertujuan untuk menguji sitotoksisitas dari senyawa ligan hidrazon berbasis turunan benzaldehida dan benzohidrasida pada Cancer Stem Cell (CSC). Analisis sitotoksisitas dilakukan dengan menggunakan metode 3-[4,5-dimetilthiazol-2yl] 2,5-difeniltetrazolium bromide (MTT Assay). Data yang diperoleh kemudian dianalisa regresi probit dengan menggunakan software SPSS Statistic, sehingga diperoleh nilai IC50. Hasil nilai IC50 dari senyawa benzolydene benzohydrazide sebesar 3,938 μM., senyawa (2-methylbenzylidene) benzohydrazide sebesar 0,145 μM, senyawa (2-nitrobenzylidene) benzohydrazide sebesar 1,319 μM, senyawa (2-bromobenzylidene) benzohydrazide) sebesar 3,398 μM dan doksorubisin sebagai kontrol positif sebesar 0,406 μM. Perbedaan nilai IC50 tersebut disebabkan karena penambahan gugus yang mampu menginduksi apoptosis.

Hydrazone compounds have potential as anticancer. Further testing is needed regarding the activity of hydrazone ligand compounds based on benzaldehyde and benzohydrasides against Cancer Stem Cells (CSC). CSCs can be used as biomarkers for early detection of tumors, determining prognosis and predicting response to therapy, while targeted therapies for CSCs can be developed into anti-cancer therapies that have the potential to cure. This study aims to examine the cytotoxicity of hydrazone ligand compounds based on benzaldehyde and benzohydraside derivatives in Cancer Stem Cells (CSC). Cytotoxicity analysis was performed using the 3-[4,5-dimethylthiazol-2yl] 2,5-diphenyltetrazolium bromide (MTT Assay) method. The result was analyzed with polynomial regression using SPSS software, so that the IC50 value was obtained. Results showed that the IC50 value of benzolydene benzohydrazide is 3,938 μM, (2-methylbenzylidene) benzohydrazide is 0,145 μM, (2-nitrobenzylidene) benzohydrazide is 1,319 μM, (2-bromobenzylidene) benzohydrazide) is 3,938 μM and doxorubicin as a positive control is 0,406 μM. The difference in IC50 value was caused by the addition of a group capable so it can induce apoptosis.
