Abstract
Energi terbarukan salah satunya biogas merupakan solusi alternatif dalam mengurangi ketergantungan terhadap bahan bakar fosil. Salah satu tahap penting dalam produksi biogas adalah hidrolisis selulosa oleh enzim selulase yang dihasilkan bakteri selulolitik pada reaktor biogas. Kondisi reaktor biogas cenderung bersifat alkali (pH ≥ 8) karena akumulasi amonia dan penyesuaian pH reaktor. Namun tidak semua bakteri selulolitik mampu bertahan dalam kondisi basa, sehingga diperlukan isolat yang toleran terhadap pH alkali sekaligus memiliki aktivitas selulolitik yang baik. Penelitian ini bertujuan untuk mengisolasi bakteri selulolitik dari slurry hasil fermentasi anaerobik biogas pada kondisi alkali, serta menguji kemampuan aktivitas selulase yang dihasilkan. Tahapan penelitian meliputi isolasi pada media selektif Carboxymethyl Cellulose (CMC) dengan variasi pH 8-10, uji skrining, pemurnian isolat, serta identifikasi makroskopis dan mikroskopis morfologi koloni dan pewarnaan Gram. Aktivitas selulase dianalisis secara kuantitatif menggunakan metode DNS untuk mengukur gula reduksi hasil hidrolisis selulosa. Selain itu, kadar protein enzim ditentukan dengan metode Bradford menggunakan reagen Coomassie Brilliant Blue G-250 untuk mengetahui konsentrasi protein total dalam ekstrak kasar selulase. Hasil penelitian menunjukkan bahwa isolat SL1 merupakan isolat dengan kemampuan selulolitik tertinggi dengan IS 3,25. Aktivitas kasar selulase tertinggi diperoleh pada pH 9 sebesar 1,734 U/mL, sedangkan aktivitas spesifik tertinggi diperoleh pada pH 10 sebesar 5,435 U/mg dominasi protein selulase target dalam ekstrak enzim. Hasil ini menunjukkan bahwa isolat SL1 berpotensi dikembangkan sebagai penghasil selulase toleran alkali untuk mendukung proses hidrolisis selulosa pada berbagai industri.

Renewable energy, such as biogas, is an alternative solution to reduce dependence on fossilfuels. One of the key stagesin biogas production isthe hydrolysis of cellulose by cellulase enzymes produced by cellulolytic bacteria in biogas reactors. Biogas reactor conditions tend to be alkaline (pH ≥ 8) due to ammonia accumulation and reactor pH adjustment. However, not all cellulolytic bacteria are able to survive under alkaline conditions; therefore, isolates that are tolerant to alkaline pH while maintaining good cellulolytic activity are required. This study aimed to isolate cellulolytic bacteria from anaerobic biogas fermentation slurry under alkaline conditions and to evaluate the cellulase activity produced. The research stages included bacterial isolation on selective Carboxymethyl Cellulose (CMC) medium at pH 8–10,screening tests, isolate purification, and macroscopic and microscopic identification of colony morphology and Gram staining. Cellulase activity was quantitatively analyzed using the DNS method to measure reducing sugars released from cellulose hydrolysis, while total protein content was determined using the Bradford method with Coomassie Brilliant Blue G-250 reagent to quantify total protein concentration in crude cellulase extracts.The results showed that isolate SL1 was the most potent cellulolytic isolate, with a cellulolytic index of 3.25. The highest crude cellulase activity was obtained at pH 9 (1.734 U/mL), while the highest specific activity was observed at pH 10 (5.435 U/mg), indicating dominance of the target cellulase protein in the enzyme extract. These findings suggest that isolate SL1 has potential to be developed as a source of alkaline-tolerant cellulase for supporting cellulose hydrolysis in various industrial applications.
