Abstract
Industri batik menghasilkan limbah cair yang mengandung pewarna naftol dan senyawa fenolik yang bersifat toksik dan sulit terdegradasi secara alami, sehingga berpotensi mencemari lingkungan. Penelitian ini bertujuan untuk menganalisis efektivitas kombinasi enzim lakase dan selobiosa dehidrogenase (CDH) dari Trametes versicolor dalam menurunkan kadar naftol, kadar total fenol, serta meningkatkan tingkat dekolorisasi pada limbah batik Jetis Sidoarjo yang terintegrasi dengan limbah domestik. Produksi enzim dilakukan melalui Submerged fermentation menggunakan substrat dedak padi untuk lakase dan tissue paper untuk CDH, diikuti dengan pemurnian parsial menggunakan presipitasi amonium sulfat dengan berbagai fraksi. Aplikasi enzim dilakukan pada limbah batik dalam sistem reaksi lakase (12 U/mL), CDH (4 U/mL), mediator ABTS (0,5 mM), dan reaksi Fenton (FeCl₃ 3 mM dan H₂O₂ 10 mM) dalam bufer natrium asetat pH 4,5 kemudian diinkubasi pada suhu ruang dengan agitasi 150 rpm selama 48 jam. Hasil uji ANOVA satu arah menunjukkan adanya perbedaan signifikan antarperlakuan terhadap efektivitas degradasi (p < 0,05). Perlakuan CDH + Fenton memberikan hasil terbaik dengan penurunan kadar naftol sebesar 77% (dari 3,10 mg/L menjadi 0,75 mg/L) dan dekolorisasi 77%. Kombinasi lakase + CDH menghasilkan penurunan kadar total fenol tertinggi sebesar 78% (dari 8,82 mg/L menjadi 1,51 mg/L), diikuti oleh lakase tunggal 67% (2,52 mg/L) dan CDH tunggal 42% (5,09 mg/L). Secara keseluruhan, kombinasi CDH + Fenton terbukti paling efektif dalam mendegradasi senyawa naftol dan lakase + CDH terbukti paling efektif dalam mendegradasi senyawa fenolik melalui mekanisme oksidasi enzimatik dan radikal hidroksil dan menunjukkan potensinya sebagai teknologi bioremediasi ramah lingkungan untuk pengolahan limbah batik berbasis pewarna sintetis.

The batik industry generates liquid waste containing naphthol dyes and phenolic compounds that are toxic and resistant to natural degradation, posing potential environmental contamination. This study aimed to analyze the effectiveness of combined laccase and cellobiose dehydrogenase (CDH) enzymes from Trametes versicolor in reducing naphthol concentration, total phenol content, and enhancing decolorization of batik wastewater from Jetis Sidoarjo integrated with domestic wastewater. Enzyme production was performed through submerged fermentation using rice bran substrate for laccase and tissue paper for CDH, followed by partial purification using ammonium sulfate precipitation at various fractions. Enzyme application was conducted on batik wastewater using a reaction system containing laccase (12 U/mL), CDH (4 U/mL), ABTS mediator (0.5 mM), and Fenton reaction (FeCl₃ 3 mM and H₂O₂ 10 mM) in sodium acetate buffer pH 4.5, then incubated at room temperature with 150 rpm agitation for 48 hours. One-way ANOVA results showed significant differences between treatments on degradation effectiveness (p < 0.05). The CDH + Fenton treatment yielded the best results with 77% reduction in naphthol concentration (from 3.10 mg/L to 0.75 mg/L) and 77% decolorization. The laccase + CDH combination achieved the highest total phenol reduction of 78% (from 8.82 mg/L to 1.51 mg/L), followed by laccase alone at 67% (2.52 mg/L) and CDH alone at 42% (5.09 mg/L). Overall, the CDH + Fenton combination proved most effective in degrading naphthol compounds, while laccase + CDH was most effective in degrading phenolic compounds through enzymatic oxidation and hydroxyl radical mechanisms, demonstrating its potential as an environmentally friendly bioremediation technology for treating batik wastewater containing synthetic dyes, particularly naphthol (azo) dyes.
