Abstract

Resistensi antimikroba merupakan permasalahan kesehatan global yang semakin meningkat dan mendorong perlunya pencarian sumber antibakteri baru dari lingkungan esktrem. Tambak garam merupakan ekosistem hipersalin yang berpotensi menjadi habitat bakteri halotoleran penghasil senyawa bioaktif. Penelitian ini bertujuan untuk menganalisis bioaktivitas antibakteri isolat bakteri halotoleran yang diiisolasi dari Tambak Garam Osowilangun, Surabaya terhadap bakteri patogen Stapyhlococcus aureus dan Escherichia coli. Isolat bakteri halotoleran (TGA 2B dan TGA 7) diregenerasi pada medai Nutrient Agar (NA) 10% dan Zobell Marine Broth (ZMB) 10%, kemudian dikarakterisasi berdasarkan morfologi koloni. Kurva pertumbuhan bakteri di analisis menggunakan pengukutan OD600 pada media NB 10% dan ZMB 10%. Uji aktivitas antibakteri dilakukan menggunakan metode difusi cakram Kiry-Bauer dengan antibiotik kontrol positi Penicillin, Gentamicin, dan Chloramphenicol. Hasil penelitian menunjukkan bahwa isolat TGA 2B dan TGA 7 mampu tumbuh pada kondisi salinitas 10% dengan karakter morfologi koloni yang bervariasi antar media. Media ZMB cenderung menghasilkan koloni yang lebih dens dan mukoid dibandingkan NA, yang mana mengindikasikan adaptasi yang lebih baik terhadap kondisi marine. Kurva pertumbuhan menunjukkan bahwa media ZMB mendukung fase log yang lebih cepat dan biomassa yang lebih tinggi pada isolat TGA 2B, sedangkan isolat TGA 7 menunjukkan fase stasioner yang lebih stabil pada media NB. Uji aktivitas antibakteri menunjukkan bahwa Penicillin menghasilkan zona hambat yang lebih konsisten terhadap S.auerus dibanding E.coli, sedangkan Gentamicin menunjukkan aktivitas terbatas dan Chloramphenicol cenderung menghasilkan zona hambat minimal atau tidak terdeteksi. Variasi respons ini menunjukkan pengaruh mekanisme kerja antibiotik, struktur dinding sel bakteri target, serta kondisi media. ===========================================================================================================
Antimicrobial resistance is an increasing global health concern that drives the need to explore new antibacterial sources from extreme environments. Salt ponds are hypersaline ecosystems that potentially serve as habitats for halotolerant bacteria capable of producing bioactive compounds. This study aimed to analyze the antibacterial bioactivity of halotolerant bacterial isolates obtained from the Osowilangun Salt Pond, Surabaya, against the pathogenic bacteria Staphylococcus aureus and Escherichia coli. Halotolerant bacterial isolates (TGA 2B and TGA 7) were regenerated on 10% Nutrient Agar (NA) and 10% Zobell Marine Broth (ZMB) and characterized based on colony morphology, while bacterial growth curves were analyzed by measuring optical density at 600 nm (OD600) in 10% Nutrient Broth (NB) and 10% ZMB. Antibacterial activity was evaluated using the Kirby–Bauer disk diffusion method with Penicillin, Gentamicin, and Chloramphenicol as positive control antibiotics. The results showed that both isolates were able to grow under 10% salinity conditions and exhibited variations in colony morphology across different media, with ZMB producing denser and more mucoid colonies, indicating better adaptation to marine-like conditions. Growth curve analysis revealed that ZMB supported a faster logarithmic phase and higher biomass production in isolate TGA 2B, whereas isolate TGA 7 showed a more stable stationary phase in NB medium. Antibacterial assays demonstrated that Penicillin produced more consistent inhibition zones against S. aureus than E. coli, while Gentamicin showed limited activity and Chloramphenicol resulted in minimal or undetectable inhibition zones, highlighting the influence of antibiotic mechanisms of action, bacterial cell wall structure, and media conditions on antibacterial responses.
