Abstract
Infeksi merupakan salah satu penyebab kematian di dunia. Sekitar 50-70% kematian di rumah sakit dilaporkan karena infeksi. Infeksi dapat menyebabkan kematian karena antibiotik yang digunakan untuk mengobati infeksi tidak dapat menghambat pertumbuhan bakteri karena resistensinya terhadap antibiotik. Mikroorganisme menghasilkan metabolit sekunder untuk mempertahankan hidup, baik mereka menggunakannya untuk memangsa atau melindungi diri mereka sendiri. Salah satu metabolit yang dihasilkan oleh mikroorganisme adalah antibiotik. Senyawa yang diperoleh dari bahan alam, terutama tumbuhan dan mikroorganisme, telah memberikan hasil yang menjanjikan dalam pengembangan senyawa antibiotik baru. Diantara jenis mikroorganisme yang ada, Actinomycetes adalah sekelompok mikroorganisme penghasil senyawa antibakteri. Tujuan penelitian ini adalah untuk untuk mengetahui aktivitas antibakteri ekstrak etil asetat isolat Actinomycetes strain AB8 yang diisolasi dari lumpur panas sidoarjo dengan menganalisis nilai kadar hambat minimumnya terhadap bakteri uji gram negatif P. aeruginosa dan E. coli dengan menggunakan metode difusi cakram Kirby-Bauer dan mikrodilusi. Isolat Actinomycetes AB8 telah diisolasi oleh Arifiyanto dkk (2020) dikirim dari Universitas Lampung ke Laboratorium Kimia Mikroorganisme Institut Teknologi Sepuluh Nopember, diregenerasi dan diuji aktivitas antagonistiknya menggunakan metode cross streak culture. Actinomycetes AB8 difermentasi selama pada suhu kamar (28-30°C) 14 hari kemudian supernatan dan biomassa dipisahkan dengan proses penyaringan. Supernatan diekstraksi dengan etil asetat (1:1 v/v) dan biomassa dimaserasi. Selanjutnya hasil ekstraksi metabolit sekunder diujikan aktivitas antibakterinya dengan metode difusi cakram Kirby-Bauer. Hasil uji terhadap bakteri gram negatif E. coli dan P. aeruginosa menunjukkan bahwa isolat Actinomycetes AB8 berpotensi menghasilkan senyawa antibakteri dengan zona hambat terbesar yaitu 215 mm pada pengujian terhadap bakteri P. aeruginosa. Selain itu, tidak terlihat aktivitas antibakteri isolate Actinomycetes AB8 pada konsentrasi 6,25 mg/mL, dimana konsentrasi tersebut merupakan konsentrasi hambat minimum (KHM) untuk melawan bakteri gram negatif E. coli dan P. aeruginosa.===================================================================================================================================
Infection is one of the causes of death in the world. Approximately 50-70% of deaths in hospital are reported due to infection. Infections can cause death because the antibiotics used to treat infections cannot inhibit the growth of bacteria due to their resistance to antibiotics. Microorganisms produce secondary metabolites to sustain life, whether they use them to prey on or protect themselves. One of the metabolites produced by microorganisms is an antibiotic. Compounds obtained from natural materials, especially plants and microorganisms, have given promising results in the development of new antibiotic compounds. Among the types of microorganisms that exist, Actinomycetes are a group of microorganisms that produce antibacterial compounds. The purpose of this study was to determine the antibacterial activity of the ethyl acetate extract of Actinomycetes strain AB8 isolated from Sidoarjo hot mud by analyzing its minimum inhibitory value against gram-negative bacteria P. aeruginosa and E. coli using the Kirby-Bauer disc diffusion method. and microdilution. Actinomycetes AB8 isolate has been isolated by Arifiyanto et al (2020) sent from the University of Lampung to the Laboratory of Microorganism Chemistry, Sepuluh Nopember Institute of Technology, regenerated and tested for antagonistic activity using the cross-streak culture method. Actinomycetes AB8 was fermented for 14 days at room temperature (28-30°C), then the supernatant and biomass were separated by a filtration process. The supernatant was extracted with ethyl acetate (1:1 v/v) and the biomass was macerated. Furthermore, the results of secondary metabolite extraction were tested for antibacterial activity by the Kirby-Bauer disk diffusion method. The test results against gram-negative bacteria E. coli and P. aeruginosa showed that Actinomycetes AB8 isolate had the potential to produce antibacterial compounds with the largest inhibition zone of 215 mm in the test against P. aeruginosa bacteria. In addition, the antibacterial activity of Actinomycetes AB8 isolate was not seen at a concentration of 6.25 mg/mL, where this concentration was the minimum inhibitory concentration (MIC) against gram-negative bacteria E. coli and P. aeruginosa.
