Abstract

Pada penelitian ini, potensi bunga kamboja jepang (Adenium obesum) sebagai antibakteri diuji terhadap E. coli dan S. aureus. Bunga kamboja jepang yang diperoleh selanjutnya diekstrak dengan metode maserasi menggunakan empat variasi pelarut (etil asetat, aseton, etanol, dan akuades). Tiap ekstrak yang diperoleh kemudian diuji kandungan fitokimia serta potensi antibakterinya terhadap E. coli dan S. aureus secara in vitro dengan metode difusi cakram dan mikrodilusi. Simplisia bunga kamboja jepang yang diekstrak menggunakan akuades menghasilkan rendemen ekstrak paling banyak (13,28%). Sedangkan simplisia bunga kamboja jepang yang diekstrak menggunakan etil asetat menghasilkan rendemen ekstrak paling sedikit (4,17%). Hasil uji kandungan fitokimia menunjukkan bahwa tiap ekstrak memiliki kandungan fitokimia beragam. Rendemen ekstraksi menggunakan pelarut etil asetat mengandung senyawa flavonoid, alkaloid, dan glikosida; rendemen ekstraksi menggunakan pelarut aseton mengandung senyawa flavonoid, alkaloid, saponin, tannin, dan glikosida; rendemen ekstraksi menggunakan pelarut etanol mengandung senyawa flavonoid, alkaloid, saponin, tannin, dan glikosida; rendemen ekstraksi menggunakan pelarut akuades mengandung senyawa flavonoid, alkaloid, dan tannin. Melalui uji antibakteri difusi cakram, rendemen ekstraksi menggunakan pelarut etil asetat memberikan penghambatan paling baik terhadap E. coli dan S. aureus dibandingkan variasi rendemen lainnya. Selanjutnya, nilai konsentrasi hambat minimum (KHM) rendemen ekstraksi menggunakan pelarut etil asetat terhadap E. coli dan S. aureus ditentukan melalui metode mikrodilusi. Untuk bakteri E. coli, nilai KHM dicapai pada konsentrasi 93,55 mg/ml. Sedangkan untuk bakteri S. aureus, nilai KHM dicapai pada konsentrasi 41,18 mg/ml.===================================================================================================================================
In this study, the potential of Adenium obesum flowers as antibacterial was tested against E. coli and S. aureus. The Adenium obesum flowers obtained were extracted by maceration using four variations of solvents (ethyl acetate, aceton, ethanol, and akuadest). Each extract obtained was tested for its phytochemical contents and its antibacterial potential against E. coli and S. aureus by disc diffusion and microdilution methods. The Adenium obesum flowers extracted using akuadest gave the highest extract yield (13,28%). Meanwhile, the Adenium obesum flowers extracted using ethyl acetate gave the least extract yield (4,17%). The results of phytochemical screening showed that each extract contained various phytochemical compounds. Extraction using ethyl acetate solvent contained flavonoid, alkaloids, and glycosides; extraction using acetone solvent contained flavonoid, alkaloids, saponins, tannins, and glycosides; extraction using ethanol solvent contained flavonoid, alkaloids, saponins, tannins, and glycosides; extraction yield using akuadest contained flavonoid, alkaloids, and tannins. Through the disc diffusion test, the extraction using ethyl acetate was the best inhibitor against E. coli and S. aureus compared to other variations. Furthermore, the value of the minimum inhibitory concentration (MIC) of the extraction using ethyl acetate solvent against E. coli and S. aureus was determined using the microdilution method. For E. coli, MIC value was reached at a concentration of 93,55 mg/ml. As for S. aureus, MIC value was reached at a concentration of 41,18 mg/ml.
