Abstract

Ganoderma boninense merupakan patogen utama penyebab penyakit Busuk Pangkal Batang pada kelapa sawit (Elaeis guineensis Jacq.), yang dapat menurunkan produktivitas tanaman secara signifikan. Penelitian ini bertujuan untuk mengetahui keanekaragaman bakteri pada batang kelapa sawit (E. guineensis Jacq.) yang terserang Ganoderma boninense berdasarkan karakter morfologi dan biokimia menurut Bergey’s Manual of Determinative Bacteriology dan gen 16S rRNA menggunakan metode NGS (Next Generation Sequencing). Sampel adalah pangkal batang kelapa sawit yang telah roboh akibat infeksi Ganoderma boninense yang diambil dari perkebunan kalapa sawit di Kota Palembang, Sumatera Selatan dan waktu roboh pohon tidak diketahui. Metode kerja pada penelitian ini diawali dengan mengisolasi bakteri pada sampel batang kelapa sawit (E. guineensis Jacq.) yang terinfeksi G. boninense, kemudian dilanjutkan dengan purifikasi bakteri. Identifikasi dilakukan pada tingkat filum dan genus berdasarkan karakteristik morfologi dan biokimia menurut Bergey’s Manual of Determinative Bacteriology, serta berdasarkan gen 16S rRNA menggunakan metode NGS. Tahapan berikutnya yaitu analisis data hasil identifikasi. Analisis data pada penelitian ini menggunakan pendekatan eksploratif deskriptif. Hasil identifikasi berdasarkan metode kultur menunjukkan bahwa bakteri yang berhasil diisolasi berasal dari tiga filum utama, yaitu Firmicutes, Actinobacteria, dan Proteobacteria. Sebanyak 19 isolat bakteri berhasil diidentifikasi dengan kecenderungan genus meliputi Bacillus, Corynebacterium, Neisseria, Vibrio, Yersinia, Acinetobacter, dan Staphylococcus, dengan genus Bacillus yang paling dominan. Sementara itu, analisis NGS menunjukkan keanekaragaman bakteri yang lebih tinggi dibandingkan metode kultur, dengan dominasi filum Pseudomonadota, Bacteroidota, dan Actinomycetota, serta ditemukannya filum lain seperti Bacillota, Acidobacteriota, dan Verrucomicrobiota. Perbedaan hasil yang diperoleh menunjukkan bahwa metode kultur hanya mampu mendeteksi bakteri yang dapat tumbuh pada media yang digunakan, sedangkan NGS mampu menggambarkan komposisi bakteri secara lebih luas berdasarkan materi genetik yang terdapat dalam sampel.

Ganoderma boninense is the primary pathogen causing Basal Stem Rot disease in oil palm (Elaeis guineensis Jacq.), which can significantly reduce crop productivity. This study aimed to determine the bacterial diversity associated with oil palm stems infected by Ganoderma boninense based on morphological and biochemical characteristics according to Bergey’s Manual of Determinative Bacteriology and 16S rRNA gene analysis using Next Generation Sequencing (NGS). The sample consisted of the basal stem of a fallen oil palm infected by G. boninense, collected from an oil palm plantation in Palembang, South Sumatra, Indonesia. The exact time at which the tree had fallen was unknown. The study was initiated by isolating bacteria from infected oil palm stem tissues, followed by bacterial purification. Identification was performed at the phylum and genus levels based on morphological and biochemical characteristics, as well as 16S rRNA gene analysis using NGS. The resulting data were analyzed using a descriptive exploratory approach. Culture-based identification revealed that the isolated bacteria belonged to three major phyla, namely Firmicutes, Actinobacteria, and Proteobacteria. A total of 19 bacterial isolates were identified with affinities to the genera Bacillus, Corynebacterium, Neisseria, Vibrio, Yersinia, Acinetobacter, and Staphylococcus, with Bacillus being the dominant genus. In contrast, NGS analysis revealed a higher bacterial diversity, dominated by the phyla Pseudomonadota, Bacteroidota and Actinomycetota, along with the presence of Bacillota, Acidobacteriota, and Verrucomicrobiota. The differences between the two methods indicate that culture-based techniques detect only bacteria capable of growing on the selected media, whereas NGS provides a broader representation of the bacterial community based on genetic material directly recovered from the sample.
