Abstract
Tanaman porang (Amorphophallus muelleri Blume) merupakan komoditas lokal bernilai ekonomi tinggi karena kandungan glukomanannya, dengan potensi pengembangan pada lahan marginal termasuk tanah masam. Porang dilaporkan mampu tumbuh pada pH rendah, namun informasi mengenai batas toleransi dan mekanisme adaptasinya terhadap cekaman masam aluminium pada tingkat morfologi, anatomi, fisiologi, dan molekuler masih terbatas. Penelitian ini bertujuan mengetahui respon morfologi, anatomi, fisiologi, dan molekuler tanaman porang asal bulbil pada cekaman masam akibat pengaruh Al₂(SO₄)₃ dengan konsentrasi 0, 5, 10, 15, dan 20 ppm. Penelitian disusun menggunakan Rancangan Acak Lengkap (RAL). Hasil penelitian menunjukkan tinggi tanaman, panjang akar, jumlah akar, dan luas daun menurun seiring meningkatnya konsentrasi, sedangkan berat dan diameter umbi batang meningkat pada konsentrasi 10 ppm kemudian menurun pada 20 ppm. Diameter epidermis akar tidak berpengaruh nyata pada seluruh perlakuan, sedangkan diameter korteks dan berkas pembuluh akar meningkat pada konsentrasi 5–10 ppm dan menurun pada konsentrasi 15–20 ppm; keempat tipe kristal kalsium oksalat (druse, rafida, prisma, stiloid) tetap terbentuk pada seluruh perlakuan. Kadar glukomanan tertinggi diperoleh pada konsentrasi 5 ppm (83,81%), kadar klorofil dan karotenoid meningkat pada konsentrasi 15–20 ppm, dan akumulasi aluminium pada akar tertinggi pada konsentrasi 10 ppm (494,21 mg/kg). Ekspresi gen SuSy2 meningkat hingga 5,15 kali pada konsentrasi 20 ppm, sedangkan ekspresi gen CSLA3 meningkat tajam hingga 2,8 kali pada konsentrasi 15 ppm dibandingkan kontrol. Pada semua perlakuan cekaman Al tanaman tumbuh dan menghasilkan umbi yang menunjukkan porang toleran terhadap cekaman masam aluminium.

Porang (Amorphophallus muelleri Blume) is a local commodity of high economic valuedue to its glucomannan content, with potential for cultivation on marginal lands, includingacidic soils. Porang has been reported to grow under low pH conditions; however, informationon its tolerance threshold and adaptation mechanisms to aluminum-induced acid stress at themorphological, anatomical, physiological, and molecular levels remains limited. This studyaimed to determine the morphological, anatomical, physiological, and molecular responses ofporang derived from bulbils to acid stress induced by Al₂(SO₄)₃ at concentrations of 0, 5, 10,15, and 20 ppm. The study was arranged using a Completely Randomized Design (CRD). Theresults showed that plant height, root length, root number, and leaf area decreased withincreasing concentration, whereas tuber weight and diameter increased at 10 ppm beforedeclining at 20 ppm. Root epidermis diameter was not significantly affected across alltreatments, whereas cortex diameter and vascular bundle diameter increased at 5–10 ppm anddecreased at 15–20 ppm; all four types of calcium oxalate crystals (druses, raphides, prisms,and styloids) were consistently formed across all treatments. The highest glucomannan contentwas obtained at 5 ppm (83.81%), chlorophyll and carotenoid content increased at 15–20 ppm,and aluminum accumulation in roots was highest at 10 ppm (494.21 mg/kg). SuSy2 geneexpression increased up to 5.15-fold at 20 ppm, while CSLA3 gene expression increasedsharply up to 2.8-fold at 15 ppm compared to the control. All plants across all treatment levelscontinued to grow and produce tubers until harvest without any mortality, indicating thatporang is tolerant to aluminum-induced acid stress.
