Abstract
Nanodispersi turunan imidazol merupakan senyawa yang berpotensi sebagai agen terapeutik, namun cenderung mengalami agregasi partikel. Penelitian ini bertujuan mengevaluasi nanodispersi turunan imidazol melalui metode represipitasi dengan memanfaatkan biopolimer sebagai coating dan glutaraldehida (GA) sebagai crosslinker. Turunan imidazol yang digunakan, yaitu 2,4,5-Triphenyl-1H-imidazole (IMD(BD)), 2-(4-Methoxyphenyl)-4,5-diphenyl-1H-imidazole (IMD(OME)), dan 2-(4-Nitrophenyl)-4,5-diphenyl-1H-imidazole (IMD(NO)). Optimasi formulasi dilakukan melalui pendekatan sequential screening pada jenis biopolimer (gelatin kakap, gelatin sapi dan pektin(PEC)), konsentrasi biopolimer serta konsentrasi GA. Karakterisasi dilakukan dengan PSA untuk mengukur ukuran partikel, indeks polidispersitas (PDI), zeta potensial, serta FESEM untuk menganalisis morfologi partikel. Selain itu, evaluasi stabilitas terhadap pH, redispersibilitas dan stabilitas penyimpanan juga dilakukan. Hasil menunjukkan IMD(NO) memiliki ukuran partikel yang lebih mendekati rentang EPR (10-200 nm) dibandingkan IMD(BD) dan IMD(OME). Pada tahap seleksi biopolimer, PEC dengan konsentrasi 25 μg/mL dipilih sebagai coating dengan ukuran partikel 206 nm dan PDI 0,353. Stabilitas redispersi diperoleh melalui penambahan GA 0,01% yang ditunjukkan oleh perubahan ukuran partikel yang relatif kecil dari 228 nm menjadi 235 nm setelah redispersi. Pada formulasi IMD(NO)-PEC-GA menunjukkan ukuran partikel 303 nm pada pH 5 dan 271 nm pada pH 7, nilai zeta potensial sebesar -23,82 mV dan mampu mempertahankan ukuran partikel yang relatif stabil hingga 14 hari penyimpanan. Berdasarkan hasil FESEM, morfologi berfaset teramati pada formulasi GA, sedangkan setelah penambahan GA diperoleh morfologi yang lebih beragam.

Imidazole derivative nanodispersion has considerable potential as a therapeutic agent, however, it is prone to particle aggregation. This study aimed to evaluate imidazole derivative nanodispersion prepared by reprecipitation method using biopolymers as coating materials and glutaraldehyde (GA) as crosslinking agent. The imidazole derivatives used were 2,4,5-Triphenyl-1H-imidazole (IMD(BD)), 2-(4-Methoxyphenyl)-4,5-diphenyl-1H-imidazole (IMD(OME)), and 2-(4-Nitrophenyl)-4,5-diphenyl-1H-imidazole (IMD(NO)). Formulation optimization was carried out using a sequential screening approach to evaluate the type of biopolymer (fish gelatin, bovine gelatin, and, pectin (PEC)), biopolymer concentration, and GA concentration. Particle size, polydispersity index (PDI), and zeta potential were characterized using particle size analysis (PSA), whereas particle morphology was examined using field emission scanning electron microscopy (FESEM). In addition, pH stability, redispersibility, and storage stability were evaluated. The results showed that IMD(NO) exhibited a smaller particle sizes to the EPR range (10-200 nm) among the evaluated imidazole derivatives. During biopolymer screening, PEC at a concentration of 25 μg/mL was selected as the coating material, as indicated by a size of 206 nm and a PDI of 0,353. Redispersibillity was improved by the addition of 0,01% GA, as indicated by only a slight increase in particle size from 228 to 235 nm after redispersion. The IMD(NO)-PEC-GA formulation exhibited particle sizes of 303 nm at pH 5 and 271 nm at pH 7, a zeta potential of -23,82 mV and maintained relatively stable particle size for up to 14 days of storage. FESEM analysis revealed faceted morphology in the formulation without GA, whereas more diverse particle morphologies were observed after the addition of GA.
