Abstract
Infeksi Staphylococcus aureus pada luka kulit menghambat penyembuhan melalui pembentukan biofilm dan perpanjangan fase inflamasi yang menekan aktivitas fibroblas, sintesis kolagen, dan angiogenesis. Penelitian ini bertujuan mengevaluasi pengaruh aplikasi topikal formulasi poliherbal SKLY1-SC10 (kombinasi Anacardium occidentale, Zingiber montanum, dan Aloe vera) terhadap ekspresi gen Col1a1, TGF-β1, dan VEGF-A pada jaringan kulit di sekitar luka Mus musculus yang terinfeksi S. aureus. Penelitian ini menggunakan mencit jantan galur DDY dengan model luka eksisi terinfeksi S. aureus USA300. Hewan uji diberi perlakuan topikal SKLY1-SC10 konsentrasi 0%, 5%, dan 10%, serta kelompok Betadine, kontrol negatif, dan kontrol positif. Sampel jaringan diambil pada hari ke-0, 7, 11, dan 14 pasca perlakuan, kemudian dianalisis menggunakan RT-qPCR (metode 2-ΔΔCt) dan dianalisis secara statistik menggunakan uji yang sesuai. Hasil menunjukkan bahwa ekspresi gen VEGF-A cenderung meningkat pada kelompok perlakuan pada beberapa waktu pengamatan, sedangkan ekspresi gen Col1a1 dan TGF-β1 berfluktuasi tanpa pola yang konsisten. Meskipun analisis statistik menunjukkan perbedaan signifikan pada ekspresi gen VEGF-A hari ke-11 dan ke-14, secara keseluruhan ekspresi gen Col1a1, TGF-β1, dan VEGF-A tidak menunjukkan perbedaan yang signifikan secara statistik antar kelompok perlakuan. Dengan demikian, aplikasi topikal formulasi poliherbal SKLY1-SC10 belum terbukti memberikan pengaruh yang signifikan terhadap ekspresi gen Col1a1, TGF-β1, dan VEGF-A pada jaringan kulit di sekitar luka Mus musculus yang terinfeksi Staphylococcus aureus.

Staphylococcus aureus infection in skin wounds impairs the wound healing process through biofilm formation and prolongation of the inflammatory phase, thereby suppressing fibroblast activity, collagen synthesis, and angiogenesis. This study aimed to evaluate the effect of the topical application of the SKLY1-SC10 polyherbal formulation (a combination of Anacardium occidentale, Zingiber montanum, and Aloe vera) on the expression of the Col1a1, TGF-β1, and VEGF-A genes in the skin tissue surrounding S. aureus-infected wounds in Mus musculus. This study used male DDY mice with an excisional wound model infected with S. aureus USA300. The animals received topical treatment with SKLY1-SC10 at concentrations of 0%, 5%, and 10%, along with Betadine, negative control, and positive control groups. Tissue samples were collected on days 0, 7, 11, and 14 post-treatment, followed by RT-qPCR analysis using the 2-ΔΔCt method and statistical analysis using appropriate statistical tests. The results showed that VEGF-A gene expression tended to increase in the treatment groups at several observation time points, whereas Col1a1 and TGF-β1 gene expression fluctuated without a consistent pattern. Although statistical analysis revealed significant differences in VEGF-A gene expression on days 11 and 14, overall Col1a1, TGF-β1, and VEGF-A gene expression did not differ significantly among the treatment groups. Therefore, the topical application of the SKLY1-SC10 polyherbal formulation has not been demonstrated to exert a significant effect on the expression of the Col1a1, TGF-β1, and VEGF-A genes in the skin tissue surrounding Staphylococcus aureus-infected wounds in Mus musculus.
