Abstract

Antosianin merupakan pigmen alami yang banyak dimanfaatkan sebagai pewarna dan indikator pH karena kemampuannya mengalami perubahan struktur dan warna akibat perubahan kondisi lingkungan. Stabilitas antosianin dipengaruhi oleh faktor-faktor seperti pH, suhu, dan cahaya sehingga evaluasi terhadap pengaruh variasi kondisi tersebut diperlukan. Penelitian ini bertujuan untuk mengevaluasi stabilitas antosianin yang diekstrak dari bunga telang (Clitoria ternatea L.) pada berbagai variasi pH, suhu, dan cahaya menggunakan metode Taguchi serta mengidentifikasi faktor yang memberikan pengaruh paling signifikan terhadap stabilitas antosianin. Ekstraksi antosianin dilakukan dengan metode Ultrasound-Assisted Extraction (UAE) menggunakan pelarut etanol-HCl 1% (v/v). Uji stabilitas disusun berdasarkan desain eksperimen Taguchi yang terdiri atas tiga faktor dan tiga level, yaitu pH (2; 4; dan 6), suhu (5; 25; dan 45 °C), serta kondisi cahaya (gelap [0 nm]; lampu LED [450 nm]; dan lampu UV-C [254 nm]). Stabilitas antosianin dievaluasi berdasarkan retensi antosianin (%) yang dihitung dari perubahan absorbansi sebelum dan sesudah perlakuan menggunakan spektrofotometer UV-Vis pada panjang gelombang maksimum. Data dianalisis menggunakan rasio Signal-to-Noise (S/N) dan Analysis of Variance (ANOVA). Hasil penelitian menunjukkan bahwa kondisi terbaik untuk mempertahankan stabilitas antosianin diperoleh pada pH 2, suhu 45 °C, dan kondisi gelap (0 nm). Hasil ini dikonfirmasi oleh hasil retensi antosianin pada kondisi terbaik yang masih berada dalam rentang interval kepercayaan 95%. Faktor-faktor yang berpengaruh signifikan terhadap stabilitas antosianin adalah pH dan suhu, sedangkan cahaya tidak berpengaruh signifikan.

Anthocyanins are natural pigments widely used as colorants and pH indicators because of their ability to undergo structural and color changes in response to changes in environmental conditions. Anthocyanin stability is influenced by factors such as pH, temperature, and light; therefore, evaluating the effects of these factors is essential. This study aimed to evaluate the stability of anthocyanins extracted from butterfly pea flowers (Clitoria ternatea L.) under various pH, temperature, and light conditions using the Taguchi method as well as to identify the factor that has the most significant effect on anthocyanin stability. Anthocyanins were extracted using the Ultrasound-Assisted Extraction (UAE) method with ethanol-HCl 1% (v/v) as the extraction solvent. The stability test was designed based on the Taguchi experimental design consisting of three factors, each at three levels: pH (2; 4; and 6), temperature (5; 25; and 45 °C), and light conditions (dark [0 nm]; LED light [450 nm]; and UV-C light [254 nm]). Anthocyanin stability was evaluated based on anthocyanin retention (%) calculated from the change in absorbance before and after treatment using a UV-Vis spectrophotometer at the maximum absorption wavelength. The data were analyzed using the Signal-to-Noise (S/N) ratio and Analysis of Variance (ANOVA). The results showed that the highest anthocyanin stability was achieved at pH 2, 45 °C, and in the dark (0 nm). These results were confirmed by the anthocyanin retention obtained under these conditions, which remained within the 95% confidence interval. Among the investigated factors, pH and temperature had significant effects on anthocyanin stability, whereas light had no significant effect.
