Abstract

Penyakit Hawar Daun Bakteri (HDB) yang disebabkan oleh Xanthomonas oryzae pv. oryzae (Xoo) merupakan salah satu penyakit penting pada tanaman padi yang dapat menurunkan produktivitas. Penelitian ini bertujuan untuk menganalisis tingkat ketahanan tujuh varietas padi, yaitu PM1, PM5, Inpari 35, Ciherang, Jeliteng, Pamerah, dan IR29 terhadap infeksi Xoo berdasarkan karakter fenotipik, fisiologis, dan molekuler. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan tiga ulangan. Inokulasi Xoo dilakukan menggunakan metode leaf clipping, kemudian diamati parameter pertambahan tinggi tanaman, jumlah anakan, panjang lesi, intensitas penyakit, kandungan klorofil relatif menggunakan SPAD, serta ekspresi relatif gen OsNPR1, OsWRKY45, dan OsPR1 menggunakan RT-qPCR. Hasil penelitian menunjukkan bahwa terdapat perbedaan respons ketahanan antar varietas terhadap infeksi Xoo. Berdasarkan keseluruhan parameter yang diamati, varietas PM5 menunjukkan tingkat ketahanan terbaik, ditandai oleh intensitas penyakit yang lebih rendah, kemampuan mempertahankan pertumbuhan dan kandungan klorofil, serta peningkatan ekspresi gen OsNPR1, OsWRKY45, dan OsPR1 setelah infeksi Xoo. Hasil penelitian ini menunjukkan bahwa ketahanan PM5 terhadap infeksi Xoo berkaitan dengan aktivasi mekanisme pertahanan yang mampu menekan perkembangan penyakit dan mempertahankan kondisi fisiologis tanaman.

Bacterial Leaf Blight (BLB), caused by Xanthomonas oryzae pv. oryzae (Xoo), is one of the major diseases affecting rice and can significantly reduce crop productivity. This study aimed to evaluate the resistance of seven rice varieties, namely PM1, PM5, Inpari 35, Ciherang, Jeliteng, Pamerah, and IR29, to Xoo infection based on phenotypic, physiological, and molecular characteristics. The experiment was arranged in a Completely Randomized Design (CRD) with three replications. Xoo inoculation was performed using the leaf-clipping method. The observed parameters included plant height increment, number of tillers, lesion length, disease intensity, relative chlorophyll content measured using a SPAD meter, and the relative expression of OsNPR1, OsWRKY45, and OsPR1 genes determined by RT-qPCR. The results showed differences in resistance responses among the rice varieties following Xoo infection. Based on the overall phenotypic, physiological, and molecular parameters, PM5 exhibited the highest level of resistance, as indicated by lower disease intensity, better maintenance of plant growth and chlorophyll content, and increased expression of OsNPR1, OsWRKY45, and OsPR1 following Xoo infection. These findings suggest that the resistance of PM5 to Xoo is associated with the activation of defense mechanisms that suppress disease development while maintaining the physiological condition of the plant.
