Abstract
Kitosan dan salep kitosan dalam penelitian ini dikembangkan dari sisik ikan Bandeng (Chanos chanos) sebagai sumber kitosan yang berasal dari limbah biomassa perikanan. Penelitian ini bertujuan mengevaluasi dan membandingkan aktivitas antibakteri larutan kitosan dan salep kitosan terhadap bakteri yang berhubungan dengan infeksi luka kulit, yaitu Staphylococcus aureus, Staphylococcus epidermidis, dan Enterococcus faecalis (bakteri Gram positif), serta Escherichia coli dan Pseudomonas aeruginosa (bakteri Gram negatif). Salep diformulasikan menggunakan basis adeps lanae dan vaselin album dengan variasi konsentrasi kitosan 5%, 10%, 15%, dan 25% serta salep betadine sebagai kontrol positif dan basis salep tanpa kitosan sebagai kontrol negatif. Pada penelitian ini, dilakukan uji antibakteri larutan kitosan dengan metode difusi cakram, uji antibakteri salep kitosan metode difusi sumuran, serta uji Minimum Inhibitory Concentration (MIC) dan Minimum Bactericidal Concentration (MBC) larutan kitosan metode mikrodilusi. Hasil penelitian menunjukkan zona hambat tertinggi pada larutan kitosan 1% dengan kategori kuat, pada bakteri Gram positif meliputi S. epidermidis (16,12 mm), E. faecalis (15,33 mm), dan S. aureus (12,83 mm), maupun bakteri Gram negatif meliputi P. aeruginosa (15,08 mm) dan E. coli (13,63 mm). Pada sediaan salep, zona hambat tertinggi dengan kategori lemah ditemukan pada konsentrasi 25% terhadap P. aeruginosa (4,25 mm) dan pada konsentrasi 5% terhadap S. epidermidis (4,25 mm). Nilai MIC berada pada rentang 625-1250 µg/mL untuk sebagian besar bakteri, sedangkan nilai MBC berkisar 1250 hingga >5000 µg/mL. Penelitian ini menunjukkan bahwa kitosan dari sisik ikan Bandeng berpotensi sebagai antibakteri topikal sekaligus memberikan pendekatan baru dalam pemanfaatan limbah perikanan untuk aplikasi biomedis.

Chitosan and chitosan ointment in this study were developed from milkfish (Chanos chanos) scales as a source of chitosan derived from fisheries biomass waste. This study aimed to evaluate and compare the antibacterial activities of chitosan solution and chitosan ointment against bacteria associated with skin wound infections, namely Staphylococcus aureus, Staphylococcus epidermidis, and Enterococcus faecalis (Gram-positive bacteria), as well as Escherichia coli and Pseudomonas aeruginosa (Gram-negative bacteria). The ointment was formulated using an adeps lanae and vaselin album base with chitosan concentrations of 5%, 10%, 15%, and 25%, with Betadine ointment as the positive control and the ointment base without chitosan as the negative control. This study included antibacterial testing of the chitosan solution using the disk diffusion method, antibacterial testing of the chitosan ointment using the agar well diffusion method, and determination of the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of the chitosan solution using the broth microdilution method. The results showed that the highest inhibition zone was produced by the 1% chitosan solution, with strong activity against Gram-positive bacteria, namely S. epidermidis (16.12 mm), E. faecalis (15.33 mm), and S. aureus (12.83 mm), as well as Gram-negative bacteria, namely P. aeruginosa (15.08 mm) and E. coli (13.63 mm). For the ointment formulation, the largest inhibition zones, categorized as weak, were observed at 25% against P. aeruginosa (4.25 mm) and at 5% against S. epidermidis (4.25 mm). MIC values ranged from 625-1250 µg/mL for most bacteria, while MBC values ranged from 1250 to >5000 µg/mL. This study shows that chitosan from milkfish scales has potential as a topical antibacterial agent while providing a new approach to utilizing fisheries waste for biomedical applications.
