Abstract

Kopi luwak liar Indonesia merupakan komoditas kopi bernilai tinggi yang berisiko mengalami pemalsuan demi mendapatkan keuntungan ekonomi yang lebih tinggi. Penelitian ini bertujuan mendiferensiasi origin kopi luwak liar Indonesia dan kopi single origin menggunakan Vis-NIR reflectance spectroscopy yang dikombinasikan dengan principal component analysis (PCA) dan hierarchical cluster analysis (HCA). Sampel kopi bubuk dari enam origin diukur pada rentang 400–1650 nm, kemudian diproses melalui baseline correction, Savitzky-Golay smoothing, MSC, dan derivative. Hasil analisis menunjukkan pola spektra kedua kelompok relatif mirip, tetapi memiliki perbedaan intensitas pada beberapa penciri kimia penting, terutama pada panjang gelombang 450–500 nm (melanoidin), 924.39 nm dan 973.78 nm (CH, CH2, dan CH3), 1244.22 nm (sukrosa dan karbohidrat), 1314.3 nm dan 1604.73 nm (kafein), 1469.96 nm (O-H, asam klorogenik, air, alkohol, dan karbohidrat), serta 1408.38 nm dan 1495.66 nm (O-H, N-H, trigonelin, asam klorogenik, kafein, dan karbohidrat). Hasil loading plot menunjukkan bahwa determinator paling berpengaruh berada pada rentang 450–500 nm melalui PC1 dan PC3, 900–950 nm melalui PC2, 1200–1350 nm melalui PC3, 1400–1550 nm melalui PC1 dan PC2, serta 1580–1630 nm melalui PC1. Hasil PCA menunjukkan adanya keterpisahan sampel pada ruang principal component, sedangkan HCA memperkuat pola kedekatan dan pengelompokan berdasarkan karakter spektra. Penelitian ini menunjukkan bahwa Vis-NIR reflectance spectroscopy yang dikombinasikan dengan PCA dan HCA dapat digunakan sebagai pendekatan cepat dan non-destruktif untuk diferensiasi kopi luwak liar Indonesia dan kopi single origin. Selain kontribusi analitis, pendekatan ini juga mendukung Sustainable Development Goals ke-8 tentang pekerjaan layak dan pertumbuhan ekonomi melalui penguatan diferensiasi kopi, perlindungan komoditas pertanian bernilai tinggi, serta pengurangan risiko praktik pemalsuan dalam rantai pasok kopi

Indonesian wild civet coffee is a high-value coffee commodity that is vulnerable to economically motivated adulteration to obtain higher profits. This study aimed to differentiate Indonesian wild civet coffee and single-origin coffee using Vis-NIR reflectance spectroscopy combined with principal component analysis (PCA) and hierarchical cluster analysis (HCA). Powdered coffee samples from six origins were measured within the 400–1650 nm wavelength range and processed using baseline correction, Savitzky-Golay smoothing, multiplicative scatter correction (MSC), and derivative transformation. The results showed that both coffee groups exhibited relatively similar spectral patterns, although distinct intensity differences were observed in several important spectral features, particularly at 450–500 nm (melanoidin), 924.39 and 973.78 nm (C-H, CH2, and CH3), 1244.22 nm (sucrose and carbohydrates), 1314.3 and 1604.73 nm (caffeine), 1469.96 nm (O-H, chlorogenic acid, water, alcohol, and carbohydrates), and 1408.38 and 1495.66 nm (O-H, N-H, trigonelline, chlorogenic acid, caffeine, and carbohydrates). The loading plot analysis indicated that the most influential differentiating regions were 450–500 nm through PC1 and PC3, 900–950 nm through PC2, 1200–1350 nm through PC3, 1400–1550 nm through PC1 and PC2, and 1580–1630 nm through PC1. PCA revealed sample separation in the principal component space, while HCA confirmed the clustering pattern based on spectral similarity. These findings demonstrate that Vis-NIR reflectance spectroscopy combined with PCA and HCA can be applied as a rapid and non-destructive approach for differentiating Indonesian wild civet coffee and single-origin coffee. In addition to its analytical contribution, this approach also supports Sustainable Development Goal 8 on Decent Work and Economic Growth by strengthening the differentiation of coffee types and origins, protecting high-value agricultural commodities, and reducing the risk of fraudulent practices in the coffee supply chain
