Abstract
Probiotik memiliki kemampuan untuk menekan pertumbuhan bakteri patogen serta menstimulasi populasi bakteri menguntungkan, sehingga berkontribusi pada pemeliharaan homeostasis mikrobioma sekum. Peningkatan populasi dan aktivitas probiotik dapat diupayakan melalui dua pendekatan, yakni suplementasi prebiotik dan makanan fermentasi. Prebiotik yang berasal dari lignocellulolisic derived oligosaccharides (LDO) dapat dihidrolisis secara enzimatik, contohnya selulase untuk meningkatkan populasi probiotik. Pemberian pakan fermentasi bakteri dan hidrolisis enzim (selulase) bertujuan untuk meningkatkan kelimpahan bakteri probiotik dan mengetahui jenis-jenisnya pada sekum mencit M. musculus Swiss Webster jantan. Respon yang diamati dari pemberian pakan berupa, kelimpahan bakteri asam laktat dengan metode TPC, dan uji biokimia untuk menentukan jenis bakteri. Hasil penelitian menunjukkan adanya peningkatan kelimpahan bakteri probiotik pada hari ke-3 di setiap perlakuan. Peningkatan kelimpahan tertinggi, yaitu pada perlakuan fermentasi bakteri hari ke-6 dengan kelimpahan sebanyak 5,73 x 108 ± 3,01 x 108 CFU/g. Hasil uji ANOVA dan Tukey HSD menunjukkan signifikansi pada perlakuan fermentasi bakteri hari ke-6. Hasil analisis keanekaragaman bakteri probiotik yang ditemukan terdapat 9 isolat, dan salah satunya dalam tingkatan jenis 1 klaster yang sama antara isolat perlakuan fermentasi bakteri dan hidrolisis enzim, seperti isolat F.5 dan E.5. Potensi probiotik dari isolat konsorsium perlakuan fermentasi bakteri memiliki aktivitas sinergis tertinggi, diikuti oleh hidrolisis enzim, dan kontrol.

Probiotics possess the ability to suppress the growth of pathogenic bacteria while stimulating the population of beneficial bacteria, thereby contributing to the maintenance of cecal microbiome homeostasis. The enhancement of probiotic populations and their activity can be achieved through two primary approaches: prebiotic supplementation and fermented feed. Prebiotics derived from lignocellulosic-derived oligosaccharides (LDO) can undergo enzymatic hydrolysis using cellulase to promote probiotic population growth. The administration of bacterial-fermented feed and enzymatically hydrolyzed feed (using cellulase) aimed to increase the abundance of probiotic bacteria and identify their species in the cecum of male Swiss Webster mice (Mus musculus). The observed parameters following feed administration included the abundance of lactic acid bacteria, quantified via the Total Plate Count (TPC) method, and biochemical assays to identify the bacterial species. The results demonstrated an increase in probiotic bacterial abundance on day 3 across all treatment groups. The highest increase was recorded in the bacterial fermentation treatment on day 6, reaching a peak abundance of 5,73 x 108 ± 3,01 x 108 CFU/g. Subsequent ANOVA and Tukey's HSD tests revealed statistical significance for the bacterial fermentation treatment on day 6. Diversity analysis of the identified probiotic bacteria yielded 9 isolates. Notably, one identical cluster was shared between the isolates from the bacterial fermentation and enzymatic hydrolysis treatments (specifically, isolates F.5 and E.5). Furthermore, the probiotic potential of the consortium isolates from the bacterial fermentation treatment demonstrated the highest synergistic activity, followed by the enzymatic hydrolysis treatment and the control group.
