Abstract
Cekaman salinitas tinggi merupakan salah satu hambatan utama dalam produksi padi (Oryza sativa L.). Penelitian ini bertujuan mengetahui respon fisiologis sembilan varietas padi lokal Indonesia (Mantap, Pamerah, Jeliteng, Situ Bagendit, Pamelen, Inpago 10, Gilirang, IR64, dan Inpari 30) terhadap cekaman NaCl (0 mM, 100 mM, dan 200 mM) menggunakan metode hidroponik pada fase vegetatif awal. Parameter yang diamati meliputi daya perkecambahan, laju pertumbuhan, vigoritas, kandungan pigmen fotosintesis, serta electrolyte leakage membran sel. Hasil uji ANOVA Two-Way menunjukkan bahwa salinitas 100 mM secara signifikan (p < 0,05) menurunkan daya perkecambahan, laju pertumbuhan, dan vigoritas seluruh varietas. Varietas Pamelen paling sensitif pada fase ini dengan penurunan laju pertumbuhan tertajam (hingga ke level 39,44), sedangkan Situ Bagendit merupakan yang paling toleran dengan daya perkecambahan stabil (>95%). Perlakuan salinitas hingga 200 mM tidak berpengaruh nyata (p > 0,05) terhadap kandungan pigmen fotosintesis (klorofil a, b, total, dan karotenoid) karena lebih dipengaruhi faktor genetik, di mana Gilirang konsisten memiliki klorofil total tertinggi (439,98 μg/ml). Sebaliknya, salinitas berpengaruh sangat nyata (p = 0,000) meningkatkan nilai electrolyte leakage. Peningkatan kerusakan membran tertinggi dialami oleh Situ Bagendit (45,12%) dan Inpago 10, sedangkan Pamelen dan IR64 lebih rendah. Sehingga tingkat toleransi padi terhadap salinitas bersifat spesifik dan bervariasi tergantung varietas serta fase parameter yang diuji.

High salinity stress is a major constraint in rice (Oryza sativa L.) production. This study aimed to evaluate the physiological responses of nine Indonesian local rice varieties (Mantap, Pamerah, Jeliteng, Situ Bagendit, Pamelen, Inpago 10, Gilirang, IR64, and Inpari 30) to NaCl stress (0 mM, 100 mM, and 200 mM) using a hydroponic method at the early vegetative stage. Observed parameters included germination percentage, growth rate, vigor, photosynthetic pigment contents, and cell membrane electrolyte leakage. Two-way ANOVA analysis revealed that 100 mM salinity significantly (p < 0.05) decreased the germination percentage, growth rate, and vigor across all varieties. Pamelen was the most sensitive variety during this stage, showing the sharpest decline in growth rate (to 39.44), whereas Situ Bagendit was the most tolerant, maintaining a stable germination percentage (>95%). Salinity treatment up to 200 mM had no significant effect (p > 0.05) on photosynthetic pigments (chlorophyll a, b, total, and carotenoids) as they were primarily governed by genetic traits, with Gilirang consistently exhibiting the highest total chlorophyll (439.98 μg/ml). Conversely, salinity stress significantly increased electrolyte leakage (p = 0.000). The highest membrane damage occurred in Situ Bagendit (45.12%) and Inpago 10, while Pamelen and IR64 showed lower increases. Therefore, salinity tolerance in rice is highly specific and varies depending on the variety and the evaluated growth parameters.
