Abstract

Kondisi salinitas tinggi menjadi hambatan utama budidaya padi di kawasan pesisir, termasuk Mangrove Gunung Anyar, Surabaya, karena mengganggu penyerapan air dan hara serta menurunkan pertumbuhan akar pada fase vegetatif. Fungi Mikoriza Arbuskula (FMA) merupakan agen hayati yang berpotensi memperbaiki morfologi akar dan meningkatkan toleransi padi terhadap cekaman salin. Penelitian ini bertujuan mengetahui pengaruh variasi dosis FMA terhadap pertumbuhan akar padi varietas Biosalin 2 pada lahan salin. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 6 taraf perlakuan dan masing-masing 12 ulangan. Hasil penelitian menunjukkan bahwa pemberian pupuk FMA memberikan pengaruh nyata (p<0,05) terhadap panjang akar, diameter akar, volume akar, berat basah akar, berat kering akar, kandungan air akar, dan biomassa total tanaman. Secara umum, perlakuan 50 gram menghasilkan pertumbuhan morfologi akar terbaik pada sebagian besar parameter yang diamati. Selain itu, analisis regresi juga menunjukkan bahwa morfologi akar memiliki hubungan positif dengan pertumbuhan vegetatif tanaman, terutama antara volume akar dengan jumlah daun dan jumlah anakan. Namun, hasil pengamatan mikroskopis menunjukkan bahwa tingkat kolonisasi FMA pada seluruh perlakuan adalah 0%, yang ditandai dengan tidak ditemukannya arbuskula, vesikel, maupun hifa internal pada akar. Kondisi tersebut diduga dipengaruhi oleh salinitas tanah yang tergolong sedang (EC 4,43 dS/m), kelembapan tanah yang sangat tinggi (98,75%), serta rendahnya kemampuan inokulum FMA beradaptasi pada kondisi lahan penelitian.

High salinity is one of the major constraints to rice cultivation in coastal areas, including the Gunung Anyar Mangrove, Surabaya, as it impairs water and nutrient uptake and inhibits root growth during the vegetative stage. Arbuscular Mycorrhizal Fungi (AMF) are beneficial soil microorganisms that have the potential to improve root morphology and enhance rice tolerance to salt stress. This study aimed to determine the effect of different AMF application rates on the root growth of Biosalin 2 rice cultivated in saline soil. The experiment employed a Completely Randomized Design (CRD) consisting of six treatment levels with twelve replications each. The results showed that AMF application had a significant effect (p<0,05) on root length, root diameter, root volume , root fresh weight , root dry weight, root water content, and total plant biomass. In general, the 50 g treatment produced the best root morphological growth across most observed parameters. Furthermore, regression analysis revealed a positive relationship between root morphology and vegetative growth, particularly between root volume and both leaf number and tiller number. However, microscopic observations showed that the AMF colonization rate was 0% in all treatments, as indicated by the absence of arbuscules, vesicles, and internal hyphae in the root tissues. This condition was presumably associated with moderate soil salinity (EC = 4.43 dS/m), extremely high soil moisture (98.75%), and the limited adaptability of the AMF inoculum to the environmental conditions of the study site.
