Abstract
Kekeringan dapat memberikan dampak buruk bagi pertumbuhan dan perkembangan tanaman tembakau (Nicotiana tabacum). Tanaman mengembangkan beragam strategi sebagai bentuk pertahanan terhadap cekaman kekeringan. Salah satu mekanisme yaitu sintesis metabolit sekunder. Oleh karena itu, pemahaman tentang regulasi genetik biosintesis metabolit sekunder dalam kaitannya dengan mekanisme adaptasi tanaman di kondisi kering sangat penting. Penelitian ini bertujuan untuk mengetahui dinamika ekspresi gen yang terlibat dalam biosintesis metabolit sekunder dari golongan isoprenoid dan alkaloid pada tanaman tembakau tipe Burley (varietas Jepon Mawar, Marakot dan MKY) dikondisi kekeringan. Ekspresi gen NtGGPPS, NtPMT, NtP5CS, dan NtEAS dari tiga varietas tembakau (var Marakot, Jepon mawar dan MKY) yang telah dikultur secara in vitro oleh Estiasih (2016) dan diberi cekaman PEG 0% dan 30%, dianalisa secara deskriptif kuantitatif menggunakan quantitative Real Time PCR (qRT-PCR). Hasil penelitian menunjukkan bahwa tingkat ekspresi gen NtGGPPS dan NtP5CS pada ketiga varietas uji yang diberi cekaman PEG 30% mengalami peningkatan dibandingkan dengan kontrol. Tingkat ekspresi dari gen NtEAS dan NtPMT mengalami penurunan pada ketiga varietas uji yang diberi cekaman PEG 30%.

Drought stress can negatively affect the growth and development of tobacco plants (Nicotiana tabacum). Plants have been developed a fascinating strategies to adapt under drought stress, including synthesis of secondary metabolites. Therefore, the knowledge of plant molecular responses that contribute to regulation of secondary metabolites production under drought stress is necessarily required. This study aims to determine the dynamics expresssion of genes involved in the biosynthesis of secondary metabolites from isoprenoids and alkaloids group in some varieties of Burley tobacco (Jepon Mawar, Marakot and MKY) under drought stress. The expression NtGGPPS, NtPMT, NtP5CS, and NtEAS of three tobacco varieties which have been previously cultured by Estiasih (2016) in MS medium and treated with 0% and 30% PEG, were quantitatively analyzed using quantitative Real Time PCR (qRT-PCR). The results showed that the expression of NtGGPPS and NtP5CS, in all tobacco varieties, increased during drought stress compared to control Whereas, the expression of NtPMT and NtEAS decreased when the three tobacco varieties were exposed to drought stress.
