Abstract
Padi merupakan salah satu komoditas pangan penting. Di Asia, padi yang memiliki pigmen unik seperti merah, ungu, hitam, coklat, kuning dan hijau yang telah banyak dibudidaya dan dikonsumsi. Padi varietas Bahbutong termasuk salah satu varietas beras merah asli Sumatera Utara yang telah dikeluarkan sejak tahun 1985 dan hanya cocok ditanam di lahan sawah saja. Saat ini belum terdapat penelitian yang melaporkan mengenai ketahanannya pada kondisi kering.
Pada penelitian ini padi varietas Bahbutong di induksi keragaman genetiknya dengan teknik iradiasi sinar gamma dosis 0 Gy, 100 Gy, 200 Gy dan 300 Gy selanjutnya biji di kecambahkan. Biji yang mampu berkecambah di subkultur pada media seleksi yang mengandung PEG 6000 dengan konsentrasi 0%, 10%, 20% dan 30%. Klon hasil seleksi yang menunjukkan ketahanan akan dikonfirmasi perbedaan karakter genetiknya dengan menggunakan teknik Random Amplified Polymorphic DNA (RAPD).
Hasil yang diperoleh dari penelitian ini adalah pemberian dosis iradiasi sinar gamma sebesar 100 Gy, 200 Gy, dan 300 Gy tidak memberikan pengaruh yang signifikan pada parameter viabilitas dan vigoritas dengan nilai p ≥ 0.05. Induksi cekaman kekeringan konsentrasi PEG 30% memberikan pengaruh signifikan (p = 0.00) pada parameter daya berkecambah hingga 0%. Sampel perlakuan iradiasi 200 Gy konsentrasi PEG 20% menunjukkan ketahanan dengan tinggi kecambah sebesar 2.75 cm dibandingkan dengan kontrol. Penggunaan primer OPA 02 diketahui paling efektif dalam menunjukkan polimorfisme DNA padi varietas Bahbutong sebanyak 9 fragmen DNA polimorfik. ========================================================= Rice  is  one  of  important  food  commodities in  the  world.  In  Asia, rice  has unique  pigments such as red, purple, black, brown, yellow and green  that  have  been  widely  cultivated  and  consumed.  Rice  varieties 
Bahbutong as one  of the  original  red rice  varieties from
North Sumatra which has been issued since 1985 but only suitable  to be planted in the field only. There  are  currently  no studies that report on its durability in 
dry conditions.
In  this  study,  the  rice  varieties  of  Bahbutong  in  the  induction  of genetic  diversity  with  gamma  ray  irradiation  technique  at  dose  0  Gy, 100  Gy,  200  Gy  and  300  Gy,  after  that  the  seeds  germinate.  Seeds  are able  to  germinate  will  be  subculture  on  a  selection  medium  containing PEG  6000 with concentrations of 0%,  10%, 20% and 30%. Clones that showed    endurance    performance    will    be    confirmed    in    genetic 
characteristics  using Random  Amplified  Polymorphic
DNA(RAPD) techniques. The result of this research is obtained that gamma ray irradiation at dose 100 Gy, 200 Gy, and 300 Gy does not give a  significant effect on viability  and  vigority  parameters  with  p  ≥  0.05.  The  induction  of drought stress at concentration of PEG 30% gave significant effect (p = 0.00) on germination up to 0%. Samples of irradiation treatment at dose 200 Gy and PEG concentration of 20% showed resistance with a sprout heigh
t of 2.75cm compared with controls. The use of OPA primer 02 is most  effective by showing  polymorphism  of  Bahbutong  rice  DNA varieties of 9 polymorphic DNA fragments.
