Abstract

Penyediaan benih bermutu merupakan permasalahan dalam
peningkatan produksi tembakau. Induksi mutasi merupakan salah satu cara untuk mendapatkan varietas unggul yang memiliki produktivitas tinggi dan kualitas yang baik. Beberapa penelitian mengenai penggunaan mutagen kimia EMS (Ethyl Methane Sulphonate) menunjukkan pengaruh terhadap perkecambahan benih dan morfologi tanaman sehingga diperlukan penelitian untuk mengetahui pengaruh konsentrasi mutagen EMS terhadap kualitas fisiologis benih dan morfologi bibit tanaman tembakau. Beberapa variasi konsentrasi EMS yang digunakan pada penelitian ini yaitu 0,1%, 0,5%, 1%, dan 1,5%. Hasil penelitian menunjukkan bahwa perbedaan konsentrasi mutagen EMS berpengaruh nyata terhadap daya berkecambah dan tidak berpengaruh nyata terhadap laju perkecambahan dan kekuatan tumbuh benih. Perlakuan EMS dengan konsentrasi 1% dan 1,5% menunjukkan penurunan daya
kecambah. Sedangkan pengamatan morfologi bibit tembakau
tidak dilakukan karena benih yang dikecambahkan tidak dapat
tumbuh menjadi bibit. Hal ini disebabkan oleh beberapa faktor lingkungan yaitu suhu, kelembaban dan media penanaman benih.

Provision of qualified seeds is a problem in increasing tobacco production. Induced mutation is the one way to obtain superior varieties which have high productivity and good guality. Several studies on the use of EMS (Ethyl Methane Sulphonate) chemical mutagen showed the effect on germination of seeds and plant morphology, so that a research was needed to find out the effect of EMS concentration on physiological quality of seed and seedling morphology of tobacco plants. Some variations of EMS concentration used in this study is 0,1%, 0,5%, and 1%.
The result of this research showed that difference of EMS
concentration had significant effect on seed germination and had no significant effect on germination rate and growth strength of seed. Treatment with 1% and 1,5% concentration of EMS showing decreased germination. At the same time, the
observation of seedling morphology can’t be done because the
seeds that have germinated can’t grow. This is due to by several environmental factor, as temperature, humidity, and seedling medium.
