Abstract

Tembakau (Nicotiana tabacum L.) merupakan jenis tanaman yang sangat di kenal di indonesia karena perannya yang cukup besar dalam bidang perekonomian. Cekaman genangan periodik merupakan salah satu faktor yang mempengaruhi pertumbuhan tanaman tembakau. Penelitian ini bertujuan untuk mengetahui respon tumbuhan dari 2 aspek yaitu fisiologis dan genetiknya terhadap cekaman genangan periodik. Aspek fisiologis yang diamati berupa laju fotosintesis, kandungan klorofil serta kadar Aktifitas Nitrat Reductase (ANR). Sedangkan aspek genetik yang diamati ialah ekspresi gen responsif cekaman genangan antara lain NtADH1 dan NtACS1. Perlakuan yang diberikan ialah cekaman genangan secara periodik yaitu dimulai dari perlakuan waterlogging dilanjutkan dengan perlakuan flooding. Hasil penelitian menunjukkan, Pada kondisi cekaman waterlogging varietas jepon emas memiliki respon terbaik terhadap parameter laju fotosintesis sebesar 103 µmol m-2 s-1, ANR sebesar 1,77 µmol/jam, kenaikan level ekspresi NtADH1dan NtACS1. Pada kondisi cekaman flooding, varietas jinten memiliki respon terbaik terhadap parameter laju fotosintesis 113,5 µmol m-2 s-1,kandungan klorofil sebesar 0,54 mg/l, ANR sebesar 2,79 µmol/jam dan Level Ekspresi NtADH1. Varietas Manilo menjadi varietas yang paling sensitif pada perlakuan waterlogging, dan varietas P951 menjadi varietas yang paling sensitif pada perlakuan flooding.
================================================================================================================== Tobacco (Nicotiana tabacum L.) is type of plant that popular in Indonesia because of their economic value industrial field. Water stress periodic is one of the factors that influence tobacco growth. This study aims to determine the physio-genetic response of tobacco. Physiological aspects observed in this study include photosynthesis rate, chlorophyll content and Nitrate Reductase Activity (ANR). While the genetic aspect observed the expression of responsive genes, such as NtADH1 and NtACS1 during water stress. The treatment given is water periodic stress that start from waterlogging treatment followed by flooding treatment. The results showed that in waterlogging stress conditions, jepon emas variety has the best response to the parameter of photosynthesis rate by 103 μmol m-2 s-1, ANR by 1.77 μmol/hr, and an increase of expression Level both genes. In flooding stress condition, jinten variety has the best response to photosynthetic rate parameter by 113.5 μmol m-2 s-1, chlorophyll content by 0.54 mg/l, ANR by 2.79 μmol/hr. Jinten variety are performed an augmentation of NtADH1 Expression Level compared with control. The most sensitive varities to waterlogging and flooding stress demonstrated by the manilo and P951 varities, respectively.
