Abstract

Padi (Oryza sativa L.) merupakan tanaman pangan pokok lebih dari separuh penduduk dunia. Padi varietas Lallodo merupakan padi lokal dataran tinggi Toraja dengan nilai produksi gabah giling tertinggi. Salah satu upaya mendapatkan varietas unggul demi memenuhi kebutuhan pangan adalah induksi mutagen kimia, salah satunya Ethyl Methane Sulphonate (EMS). Tujuan dari penelitian ini adalah untuk mempelajari efek EMS pada morfologi akar. Parameter yang diamati pada penelitian ini adalah daya kecambah, panjang akar dan bentuk rambut akar kecambah padi Lallodo yang dipapar dengan larutan EMS pada konsentrasi 0%; 0.25%; 0.5%; 0.75%; 1%; 1.25%; 1.5%, dan 2%. Selanjutnya nilai daya kecambah dan panjang akar dianalisis dengan menggunakan ANOVA oneway dengan dilanjutkan uji Duncan. Hasil penelitian menunjukkan bahwa perlakuan EMS berpengaruh terhadap panjang akar kecambah padi dimana perlakuan EMS 0%; 0,25%; dan 0,50% mempengaruhi panjang akar secara tidak signifikan. Perlakuan EMS juga berpengaruh pada daya kecambah pada hari 1 hingga 3, tetapi tidak berpengaruh pada daya kecambah hari 4. Pengamatan visual morfologi rambut akar dengan mikroskop dan SEM (Scanning Electron Microscope) menunjukkan bahwa semakin tinggi konsentrasi EMS maka rambut akar semakin pendek dan kerapatannya semakin jarang.

Rice (Oryza sativa L.) is the most important food crop which is a staple food of more than half of the world's population. The Lallodo variety rice is a local Toraja highland rice which has the highest yield of milled grain. To meet the increasing food needs, it is necessary to increase productivity which can be achieved through increasing genetic variation using chemical mutagens, one of which is Ethyl Methane Sulphonate (EMS). The purpose of this study was to study the effects of EMS immersion on germination, root length and root hair shape. The parameters observed in this study were germination, root length and sprout root hair form from Lallodo rice seeds exposed to EMS solutions at concentrations of 0%, 0.25%, 0.5%, 0.75%, 1%, 1.25%, 1.5%, and 2%. Furthermore, the value of sprout power and root length were analyzed using oneway ANOVA followed by Duncan test. This present study showed that EMS treatment affects the root length of rice germ, which 0%; 0,25%; and 0,50% EMS treatment does not affects significantly. EMS treatment also affects the germination rate on day one untill day three. However, it does not affects the germination on day four treatment. The shape of root hair is known by microscopic observation and using Scanning Electron Microscopy (SEM) and shows that the higher the EMS concentration, the shorter the hair shape of the root and the less frequent its density.
