Abstract

Kapsul berbahan dasar kitosan larut air (WSC) telah disintesis dengan penambahan alginat serta dengan dan tanpa penambahan surfaktan Sodium Dodesil Sulfat (SDS). Kapsul dengan dan tanpa penambahan SDS dibuat dengan variasi perbandingan WSC:Alginat yaitu 2:1, 3:1, 4:1, 5:1 dan 6:1 (w/w). Kitosan yang digunakan pada penelitian ini adalah kitosan komersial yang berasal dari cangkang rajungan. Kitosan didepolimerisasi menjadi kitosan larut air melalui metode hidrolisis dengan H2O2. Karakterisasi yang dilakukan meliputi uji FTIR, keseragaman bobot, swelling, degradasi, kelarutan dan rilis obat. Spektra FTIR pada kapsul menunjukkan adanya vibrasi dari molekul-molekul penyusun kitosan, alginat dan SDS. Hasil uji keseragaman bobot dan uji kelarutan menunjukkan bahwa kapsul yang terbentuk memenuhi syarat yang ditetapkan oleh Farmakope Indonesia. Derajat swelling terbaik dalam air dan larutan HCl 0,1 N masing-masing sebesar 84,45% dan 82,50%. Persentase degradasi terbaik dalam air dan larutan HCl 0,1 N masing-masing sebesar 97,37% dan 85,48%. Uji rilis obat terbaik dalam air dan larutan HCl 0,1 N masing-masing adalah 23 detik dan 1 menit 6 detik. Kapsul yang memiliki penampakan fisik atau tekstur yang paling baik dan mendekati kapsul komersial adalah kapsul WSC:Alginat 2:1.

Capsules made from water soluble chitosan (WSC) have been synthesized by the addition of alginate and with and without the addition of Sodium Dodecyl Sulfate (SDS) surfactant. Capsules with and without the addition of SDS are made with variations in the ratio of WSC: Alginate, i.e 2:1, 3:1, 4:1, 5:1 and 6:1 (w/w). Chitosan was used in this research is a commercial chitosan from the crab shell. Chitosan was depolymerized into water soluble chitosan by hydrolysis method with H2O2. Characterization was performed by FTIR, weight uniformity, swelling, degradation, solubility and drug release test. FTIR spektra showed that the obtained capsules had vibration from its constituent molecules, i.e chitosan, alginate and SDS. The results of the weight uniformity test and solubility test indicate that the capsules were obtained were eligible the requirements set by the Indonesian Pharmacopoeia. The best degree of swelling in water and 0,1 N HCl solution were 84,45% and 82,50% respectively. The best percentage of degradation in water and 0,1 N HCl solution were 97,37% and 85,48% respectively. The best drug release test in water and 0,1 N HCl solution were 23 seconds and 1 minute 6 seconds respectively. Capsules that have the best physical appearance or texture and are close to commercial capsules are WSC:Alginate 2:1.
