Abstract
Imobilisasi adalah teknik untuk membawa Bacillus sp.
SLII-1 ke usus unggas. Bacillus sp. SLII-1 sebagai bakteri keratinolitik diharapkan dapat meningkatkan kecernaan pakan berbahan tepung bulu. Tujuan dari penelitian ini adalah untuk menentukan konsentrasi natrium alginat paling efektif sebagai bahan imobilisasi Bacillus sp. SLII-1 dan memformulasikan pakan mengandung tepung bulu untuk burung puyuh (Coturnix japonica). Imobilisasi dilakukan menggunakan natrium alginat dilapisi kitosan dengan metode ekstrusi. Hasil imobilisasi diuji pada simulasi pencernaan unggas dengan melakukan perhitungan bakteri menggunakan metode Total Plate Count (TPC). Formulasi pakan diaplikasikan pada burung puyuh umur 12 sampai 30 hari dan diukur peningkatan berat badan dan Feed Conversion Ratio (FCR). Hasil penelitian menunjukkan bahwa konsentrasi natrium alginat paling efektif untuk imobilisasi Bacillus sp. SLII-1 adalah 2% (b/v) dengan jumlah sel terimobilisasi sebesar (4,45±0,21)x107CFU/g dan melepaskan sebesar (2,54±0,04)x107CFU/g di usus pada simulasi pencernaan unggas. Formulasi pakan VII (95% pakan standar+5% bulu+10 g suplemen Bacillus sp. SLII-1/kg pakan) menghasilkan peningkatan berat badan terbesar (5,61±0,14 g/hari) dan nilai FCR terkecil (2,80±0,11) pada burung puyuh umur 16 sampai 30 hari.

Immobilization is a technique for delivering Bacillus sp. SLII-1 to the poultry intestine. Bacillus sp. SLII-1, as a keratinolytic bacterium, is expected to improve the digestibility of feather meal-based feed. The purpose of this study was to determine the most effective concentration of sodium alginate as an immobilization matrix for Bacillus sp. SLII-1 and to formulate feather meal-based feed for quails (Coturnix japonica). Immobilization was carried out using chitosan-coated sodium alginate through the extrusion method. The immobilized bacteria were evaluated using a simulated poultry digestive system by determining bacterial counts with the Total Plate Count (TPC) method. Feed formulations were then applied to quails aged 12–30 days, and body weight gain as well as Feed Conversion Ratio (FCR) were measured. The results showed that the most effective sodium alginate concentration for immobilizing Bacillus sp. SLII-1 was 2% (w/v), with the number of immobilized cells reaching (4.45 ± 0.21) × 10^7 CFU/g and released cells reaching (2.54 ± 0.04) × 10^7 CFU/g in the intestinal phase of the poultry digestive system simulation. Formulation VII (95% standard feed + 5% feather meal + 10 g Bacillus sp. SLII-1 supplement/kg feed) resulted in the highest body weight gain (5.61 ± 0.14 g/day) and the lowest FCR value (2.80 ± 0.11) in quails aged 16–30 days.
