Abstract
Penyakit pada IVD (Intervertebral Disc) yang sering terjadi akibat kerusakan jaringan manusia yang disebabkan oleh banyak hal seperti bertambahnya usia, penyakit tertentu, posisi duduk yang kurang tepat dan juga kecelakaan pada tulang punggung. Pengobatan paling umum adalah dengan melakukan pembedahan (decompresi). Proses decompresi yang dilakukan yakni sendi tulang belakang dipotong dan diganti dengan material yang baru. Material Chitosan memiliki sifat non-toksisitas, biokompatibilitas, dan aktivitas antibakteri namun mudah untuk larut dalam cairan. Sedangkan material silicone rubber memiliki sifat inert yang artinya tidak beraksi pada tubuh saat dijadikan material implant, tahan terhadap bahan kimia dan panas. Silicone rubber RTV 585 memiliki sifat bioinert namun untuk penelitian mengenai silicone rubber RTV 585 masih terbilang sedikit. Penelitian ini bertujuan untuk menganalisa pengaruh sifat biokompatibel terhadap penggabungan antara silicone rubber dengan chitosan dan diharapkan memiliki sifat bioinert, antibakteri dan tidak mudah larut. Varisasi penambahan chitosan yang digunakan adalah 10, 20, 30, 40, dan 50% dalam volume. selanjutnya Sampel kemudian dikarakterisasi menggunakan, SEM, MTT assay, antibakteri dan hidrolitik degradasi material. Dari pengujian tersebut didapatkan penambahan chitosan akan menambah pori pada silicone rubber RTV 585, menambah diameter zona inhibisi antibakteri, dan kemampuan menumbuhkan sel fibroblas. Penambahan 50% chitosan mencatatkan nilai tertinggi pada area pori sebesar 29,282%, komposisi nitorgen sebesar 01,56% wt, memiliki zona inhibisi paling lebar sebesar 6.4± 0.4 mm, memiliki %cell viability paling banyak sebesar 80,08± 1.21%, memiliki %weight loss paling tinggi sebesar pada setiap minggunya serta lifetime paling pendek sekitar 36 minggu. Hasil pengujian dari komposit ini diharapkan dapat menjadi kandidat injectable material yang memiliki sifat biokompatibilitas dan degradasi hidrolitik yang baik.

The IVD’s (Intervertebral Disc) disease which often occurs due to damaging human tissue caused by many things such as increasing age, certain diseases, improper sitting position and also accidents on the spine. The most common treatment is surgery (decompression). The decompression itself is a process to cut and replaced the spinal joint with some new material. Chitosan material has some character which is non-toxicity, biocompatibility and antibacterial activities but so easy to dissolve in liquids. While silicone rubber material (RTV 585) has inert’s character which means it doesn’t act on the body when used as implant material, resist to chemicals and heat. This study aims to analyze the effect of biocompatible properties on the incorporation of silicone rubber with chitosan and is expected to be bioinert, antibacterial and not soluble. The variability of chitosan additions used are 10, 20, 30, 40, and 50% in volume, then the samples were characterized using, SEM, MTT assay, antibacterial and hydrolytic material degradation for a month. From this test, the addition of chitosan will increase the pore on the silicone rubber RTV 585, increases the antibacterial inhibition zone, and the ability to grow fibroblast cells. The addition of 50% chitosan recorded the highest value in the pore area of 29.282%, the composition of nitrogen was 01.56% wt, the widest zone of inhibition of 6.4 ± 0.4 mm, the highest% cell viability of 80.08 ± 1.21%, had% the highest weight loss in each week and the shortest lifetime around 36 weeks. The final results of these composites are expected to be candidates for injectable material which has good biocompatibility and hydrolytic degradation.
