Abstract
Pengobatan penyakit kanker payudara saat ini masih menggunakan agen kemoterapi seperti Doxorubicin yang menimbulkan banyak efek samping bagi sel normal. Karena itu saat ini dikembangkan senyawa dari sintesis bahan alam seperti senyawa Trisindolina 1 yang berpotensi sebagai senyawa alternatif. Penelitian ini bertujuan untuk menguji sitotoksisitas dan pengaruh senyawa Trisindolina 1 terhadap aktivitas penghambatan fase siklus sel kanker payudara MCF – 7 Cell Line. Uji sitotoksisitas senyawa dilakukan dengan uji MTT menggunakan 7 variasi konsentrasi (1,5625; 3,125; 6,25; 12,5; 25; 50; 100 μg/ ml) dan Doxorubicin sebagai kontrol untuk mendapatkan nilai IC50. Sedangkan, pengaruh senyawa Trisindolina 1 terhadap penghambatan fase siklus sel kanker MCF 7 diuji menggunakan metode flowcytometer dengan menggunakan konsentrasi 2IC50, IC50 dan ½ IC50. Sebagai kontrol positif digunakan Doxorubicin dengan konsentrasi IC50. Hasil IC50 senyawa Trisindolina 1 dan Doxorubicin secara berturut – turut sebesar 15.77 μg/ml dan 20,61 μg/ml, serta senyawa Trisindolina 1 dan Doxorubicin secara berturut – turut diketahui menyebabkan siklus sel arrest pada fase S dan fase G0/G1. ================================================================================================================================
The treatment of breast cancer is currently using chemotherapy agents such as Doxorubicin which causes many side effects for normal cells. Therefore, a compound form natural source is currently being developed, one of them is Trisindoline 1 which has the potential as alternative compound. The aims of this study are to examine the cytotoxic activity and to examine the effect of Trisindoline 1 against the cell cycle of breast cancer MCF – 7 Cell line. The MTT – Assay is used to examine the cytotoxic effect of Trisindoline 1 using 7 variations of concentration (100; 50; 25; 12,5; 6,25; 3,125; 1,525 μg/ml) and Doxorubicin as positive control to obtain the IC50 value. Meanwhile, the effect of Trisindoline 1 on the inhibition cell cycle phase of the MCF 7 was tested using the flowcytometry method using the 2IC50, IC50 and ½ IC50 concentration of Trisindoline 1. As a positive control, the IC50 concentration of Doxorubicin was used. IC50 results of Trisindoline 1 and Doxorubicin compounds were 15.77 μg/ml and 20.61 μg/ml respectively. As well as Trisindoline 1 is known to cause cell cycle arrest in the S phase, meanwhile, Doxorubicin caused cell arrest in G0/G1 phase.
