Abstract
Skeletonema costatum merupakan salah satu kelompok mikroalga yang dapat digunakan sebagai bahan baku biodiesel karena memiliki kandungan lipid dan waktu penggandaan yang relatif cepat, yaitu 0,340 hari. Akumulasi lipid mikroalga mempunyai kecenderungan untuk meningkat dalam kondisi tercekam. Penelitian ini bertujuan untuk mengetahui pengaruh fotoperiode dan cekaman fisiologis nutrien N dan Si terhadap pertumbuhan dan kandungan lipid S. costatum. Metode yang digunakan adalah kultur S. costatum pada media fisiologis cekaman nutrien N dan Si dengan variasi fotoperiode, pengukuran kepadatan sel setiap 6 jam sekali menggunakan Haemocytometer Improved Neubauer, pemanenan dilakukan di akhir fase eksponensial, lalu dilakukan perhitungan biomassa, analisis kualitatif dan analisis kuantitatif kandungan lipid. Analisis data menggunakan ANOVA two way. Hasil penelitian menunjukkan bahwa perlakuan fotoperiode, nutrien dan interaksi keduanya tidak berpengaruh terhadap biomassa S. costatum. Pertumbuhan tertinggi terdapat pada perlakuan A24 dengan kepadatan sel sebesar 132.500 sel/ml dan biomassa 0,49 gram, sedangkan kandungan lipid tertinggi terdapat pada perlakuan B24 dengan kandungan lipid sebesar 14,42%.

Skeletonema costatum is a species of microalgae with significant potential as a biodiesel feedstock due to its high lipid content and relatively short doubling time of 0.340 days. Lipid accumulation in microalgae generally increases under stress conditions. This study aimed to investigate the effects of photoperiod and physiological stress induced by nitrogen (N) and silicon (Si) nutrient limitation on the growth and lipid content of *S. costatum*. The microalgae were cultured under nitrogen and silicon nutrient stress with different photoperiod treatments. Cell density was measured every 6 hours using an Improved Neubauer hemocytometer. Harvesting was conducted at the end of the exponential growth phase, followed by biomass determination and qualitative and quantitative analyses of lipid content. Data were analyzed using a two-way analysis of variance (ANOVA). The results indicated that photoperiod, nutrient treatments, and their interaction had no significant effect on the biomass of *S. costatum*. The highest growth was obtained in treatment A24, with a cell density of 132,500 cells/mL and a biomass of 0.49 g, whereas the highest lipid content was observed in treatment B24, reaching 14.42%.
