Abstract

Sumenep dan Sampang merupakan dua kabupaten utama penghasil kacang hijau di Jawa Timur, masing-masing dengan luas tanam 11.718 ha dan 11.663 ha. Aksesi lokal yang ditanam di Sumenep adalah aksesi Manding dan aksesi Lenteng, yang mana informasi genetik dari kedua aksesi tersebut masih belum diketahui. Berdasarkan uraian di atas tujuan penelitian ini adalah mengetahui keragaman genetik kacang hijau aksesi lokal Sumenep berdasarkan marka SSR dan marka RAPD. Penelitian ini dilakukan dengan tahapan-tahapan meliputi uji viabilitas biji, isolasi DNA, kuantifikasi DNA menggunakan nanodrop, amplifikasi DNA menggunakan marka SSR dan RAPD, dan visualisasi DNA menggunakan gel elektroforesis. Data yang diperoleh dianalisis menggunakan deskriptif kuantitatif. Hasil penelitian menunjukkan bahwa, marka RAPD dan SSR dapat digunakan dalam analisis variasi genetik aksesi Lenteng dan Manding tanaman kacang hijau. Terdapat dua marka RAPD dan empat marka SSR yang masuk kategori marka cukup informatif dengan nilai PIC 0.5.

Sumenep and Sampang are the two main districts producing green beans in East Java, each with 11,718 ha and 11,663 ha. Local accession located in Sumenep is Manding accession and Lenteng accession, where information originating from both accesses is still unknown. The following is a description of the objectives of this study to determine the genetic diversity of green beans in local sumenep accession based on SSR markers and RAPD markers. This research was carried out by stages consisting of seed viability, isolation DNA, DNA quantification using nanodrop, DNA amplification use SSR and RAPD markers, and DNA visualization using gel electrophoresis. The data will be analized with quantitative descriptive. The results showed that the RAPD and SSR markers could be used in the analysis of genetic variation of the Lantern and Manding accessions of green bean plants. There were two RAPD markers and four SSR markers that were included in the markers category quite informative with a PIC 0.5 value.
