Abstract
Herbisida atrazin merupakan jenis pestisida yang paling banyak digunakan dan bersifat persisten pada tanah. Hal ini menjadikan atrazin sebagai ancaman lingkungan karena dapat mencemari tanah dan air. Yeast sebagai agen pendegradasi memiliki banyak kelebihan disebabkan toleransinya terhadap kondisi ekstrim serta memiliki respon baik terhadap senyawa beracun. Tujuan dari penelitian ini adalah mengetahui kemampuan isolat yeast dalam mendegradasi atrazin.
Isolat yang digunakan adalah tiga isolat terbaik hasil skrining melalui pengukuran zona bening, yaitu TB1, UF5, dan UF7. Ketiga isolat dikarakterisasi dan diidentifikasi berdasarkan morfologi serta fisiologi biokimia. Ketiga isolat dan kontrol dikultur dalam AMS cair 120 μg/L (berdasarkan uji respon dosis). Persentase degradasi atrazin diuji menggunakan metode spektrofotometri dan viabilitas sel yeast menggunakan metode TPC. Hasil penelitian menunjukkan bahwa semua isolat terskrining termasuk ke dalam genus Candida. Ketiga isolat mampu mendegradasi atrazin hingga 94,26% (TB1), 64,01% (UF5), dan 52,51% (UF7) dengan TPC berturut-turut sebesar 9,27x104, 7,83x104, dan 6,27x104 CFU/mL pada hari ke-10.

Atrazine herbicides are the most widely used pesticides in the world and are persistent on the ground. This makes atrazine an environmental concern because it can pollute the soil and water. Yeast as a degrading agent has many advantages because of its tolerance to extreme conditions and has a good response to toxic compounds. The purpose of this study was to determine the ability of yeast isolates to degrade atrazine.
The isolates used were the best three isolates resulting from screening through clear zone measurements, namely TB1, UF5, and UF7. The three isolates were characterized and identified based on morphology and biochemical physiology. The three isolates and control were cultured in liquid AMS 120 μg/L (based on the dose-response test). The percentage of atrazine degradation was tested using spectrophotometric methods and yeast cell viability using the TPC method. The results showed that all screened isolates belonged to the genus Candida. The three isolates were able to degrade atrazine up to 94.26% (TB1), 64.01% (UF5), and 52.51% (UF7) with TPC respectively at 9.27x104, 7.83x104, and 6.27x104 CFU/mL on the 10th day.
