Abstract
Polietilen sulit didegradasi karena tersusun atas monomer alkana yang memiliki ikatan C-C dan C-R yang bersifat stabil. Jamur Aspergillus terreus diketahui menghasilkan Alkana hidroksilase (AH), salah satu enzim yang terlibat dalam degradasi polietilen. Enzim AH membutuhkan kofaktor berupa ion Fe2+. Tujuan dari penelitian ini, untuk mengetahui pengaruh konsentrasi ion Fe2+ dan pH terhadap potensi A. terreus dalam mendegradasi plastik polietilen selama 30 hari. Penelitian ini menggunakan medium MSM dengan variasi konsentrasi ion Fe2+ dan pH. Parameter yang diamati adalah presentase degradasi dengan menggunakan timbangan analitik, pengukuran aktivitas enzim AH dengan menggunakan metode spetrofotometri, verifikasi enzim AH dilakukan dengan metode SDS-PAGE, dan analisis perubahan gugus fungsi akan dilakukan dengan menggunakan metode FTIR. Hasil menunjukkan bahwa perlakuan konsentrasi ion Fe2+ dan pH berpengaruh terhadap presentase degradasi dan aktivitas AH. Hasil tertinggi dicapai pada konsentrasi 1 ppm ion Fe2+ dan pH 7, yaitu persentase degradasi sebesar 9,4% dan aktivitas AH 11 U/mL. Pada perlakuan tersebut, hasil FTIR menunjukkan adanya peningkatan persentase transmisi di gugus C-H pada panjang gelombang 2849,5 cm-1 dan 1462,9 cm-1. Analisis SDS-PAGE juga menunjukkan adanya pita protein berukuran sekitar 46kDa, yang merupakan berat molekul dari enzim AH.

Polyethylene is difficult to degrade because it is composed of alkane monomers that have stable C-C and C-R bonds. Fungi Aspergillus terreus have Alkane hydroxilase (AH), one of the enzymes involved in the polyethylene degradation. AH requires an ion Fe2+ cofactor. The purpose of this study was to determine the effect of ion Fe2+ and pH on polyethylene degradation performed by A. terreus for 30 days. A Minimal Salt Medium was used by adding different ion Fe2+ concentration and pH. The parameters were the lost of dry weight using analytical scales, AH protein detection by SDS-PAGE, AH activity using the spetrofotometer method, and functional group changing by FTIR method. The results showed that ion Fe2+ and pH had an effect on those parameters. The highest yield was achieved at 1 ppm ion Fe2+ and and pH 7, with approximately 9,4% plastic degradation and 11 U/mL AH activity. FTIR showed increasing transmission peacks at wavelengths of 2849.5 cm-1 and 1462,9 cm-1 which were belonged to C-H. SDS-PAGE also showed one band with molecular weight around 46kDa, which was similar to the size of AH protein.
